一起急性腹瀉疫情中檢出的GI.8型諾如病毒的序列測(cè)定和分子特征分析

【摘要】:目的對(duì)湖州地區(qū)一起急性腹瀉疫情中檢出的GI.8型諾如病毒進(jìn)行序列測(cè)定和分子特征分析。方法根據(jù)GI型諾如病毒保守序列自行設(shè)計(jì)5對(duì)引物,用一步法RT-PCR分段擴(kuò)增GI.8型諾如病毒CHN/Huzhou/N10的全基因組序列,采用生物信息學(xué)相關(guān)軟件對(duì)序列進(jìn)行整理和分析。結(jié)果 CHN/Huzhou/N10全長(zhǎng)7 740bp,編碼區(qū)包括3個(gè)開(kāi)放讀碼框架(ORF1~ORF3),分別長(zhǎng)5 400bp(5~5 404nt)、1 632bp(5 388~7 019nt)和642bp(7 019~7 660nt)。RdRp區(qū),VP1區(qū)和VP2區(qū)的系統(tǒng)進(jìn)化分析顯示CHN/Huzhou/N10屬于GI.8基因型。衣殼蛋白區(qū)的氨基酸序列分析表明,與原型株Boxer/2001/US相比,CHN/Huzhou/N10VP1區(qū)的氨基酸序列共有16個(gè)變異位點(diǎn),12個(gè)位于P2區(qū)。其中aa347T→S,aa397T→E這2個(gè)變異位點(diǎn)分別位于HBGA受體結(jié)合袋位點(diǎn)Ⅱ和Ⅲ內(nèi)。結(jié)論本文獲得了我國(guó)GI.8型諾如病毒CHN/Huzhou/N10的全基因組序列,相關(guān)序列信息可用于病毒的遺傳進(jìn)化研究、快速診斷試劑的研發(fā)以及疫苗的設(shè)計(jì)。
[Abstract]:Objective to determine the sequence and molecular characteristics of GI.8 type Norovirus detected in a case of acute diarrhea in Huzhou area. Methods five pairs of primers were designed according to the conserved sequence of GI type Norovirus. The whole genome sequence of GI.8 type Norovirus CHN/Huzhou/N10 was amplified by one-step RT-PCR. The sequence was sequenced and analyzed by bioinformatics software. Results the length of CHN/Huzhou/N10 was 7 740 BP. The coding region consists of three open reading frames ORF1 / ORF3 with a length of 5 400 BP / 5 404nt respectively. The region of 1 632bpmPU (5 388n) and 642 BP (7 019nt) and 7660nt (RdRp). Phylogenetic analysis of VP1 region and VP2 region showed that CHN/Huzhou/N10 belonged to GI.8 genotype. Amino acid sequence analysis of capsid protein region showed that CHN/Huzhou/N10 belonged to GI.8 genotype. Compared with the prototype strain Boxer/2001/US, the amino acid sequence of CHN / Huzhou-N10VP1 region had 16 mutation sites. 12 are located in P2 region, of which aa347T. 鈫扴,aa397T. 鈫扙. these two mutation sites are located in the HBGA receptor binding pouch site 鈪,
