【摘要】：目的調(diào)查獻(xiàn)血人員外周血白細(xì)胞中8種人皰疹病毒的DNA分布情況,為我國獻(xiàn)血者健康標(biāo)準(zhǔn)要求的完善提供參考性實(shí)驗(yàn)數(shù)據(jù)。方法收集獻(xiàn)血人員外周血標(biāo)本427例,分離外周血白細(xì)胞后提取DNA,使用Taqman探針法熒光定量PCR檢測方法對8種皰疹病毒DNA的陽性率和病毒載量進(jìn)行檢測。結(jié)果 427例標(biāo)本中未檢測到單純皰疹病毒1型(HSV1)、HSV2、水痘-帶狀皰疹病毒(VZV)和人皰疹病毒8型(HHV-8)的DNA,EB病毒(EBV)、人巨細(xì)胞病毒(HCMV)、HHV-6、HHV-7陽性率分別為51.4%、6.5%、50.6%、21.1%,其中HCMV、EBV和HHV-6均陽性的標(biāo)本占6.5%,EBV和HHV-6均陽性的標(biāo)本占25.1%,EBV與HHV-7均陽性的標(biāo)本占4.0%,HHV-6和HHV-7均陽性的標(biāo)本占6.9%。EBV、HCMV、HHV-6、HHV-7的病毒載量中位數(shù)分別為559、336、4 683、1 126 gqe/ml外周血。在不同性別和年齡組中EBV、HHV-6和HHV-7在三個(gè)年齡組中的檢出率差異無統(tǒng)計(jì)學(xué)意義,但CMV的檢出率隨年齡的增加逐漸升高,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。市區(qū)與郊區(qū)人群相比EBV、HCMV、HHV-6和HHV-7的檢出率均顯著較低(OR=0.657、0.798、0.889、0.721,95%CI:0.147~2.451、0.224~2.196、0.221~2.703、0.194~2.445,P0.05)。結(jié)論在獻(xiàn)血人員外周血白細(xì)胞中可以檢出多種皰疹病毒DNA,提示我國獻(xiàn)血者健康標(biāo)準(zhǔn)要適時(shí)考慮獻(xiàn)血者皰疹病毒感染的情況。
[Abstract]:Objective to investigate the DNA distribution of eight human herpesviruses in peripheral white blood cells (PBWs) of blood donors. Methods the peripheral blood samples of 427 blood donors were collected and DNA was extracted from peripheral white blood cells. The positive rate and viral load of 8 herpesvirus DNA were detected by Taqman probe fluorescence quantitative PCR method. Results No herpes simplex virus type 1 was detected in 427 samples. HSV1). The DNA of HSV2, varicella-zoster virus (VZV) and human herpesvirus 8 (HHV-8) were detected by EBV. The positive rate of HHV-7 was 51.4 / 6.5 and 50.6 / 21.1respectively, of which 6.5% were positive for both HHV-7 and HHV-6. The positive specimens of EBV and HHV-6 were found to be positive for EBV and HHV-7 in 25.1. the positive specimens of HHV-6 and HHV-7 were 6.9. EBV. The median viral load of HHV-6 and HHV-7 of HCMV was 559 ~ 336n ~ 4 683, respectively. 1 126 gqe/ml peripheral blood. There was no significant difference in the detection rate of EBV HHV-6 and HHV-7 among the three age groups in different sex and age groups. But the detection rate of CMV gradually increased with the increase of age, the difference was statistically significant (P 0.05). The detectable rates of HHV-6 and HHV-7 were significantly lower than that of ORX 0.657U 0.798U 0.889 / 0.721 / 95 CI: 0.147 / 2.451. 0.224c2.196 / 0.221 / 2.703 / 0.194 / 2.445. Conclusion A variety of herpesvirus DNA can be detected in peripheral white blood cells of blood donors, which suggests that the situation of herpesvirus infection in blood donors should be taken into account in the health standard of blood donors in China.
