【摘要】：目的:近年來(lái)精子DNA碎片預(yù)測(cè)男性生育受到廣泛關(guān)注,其檢測(cè)方法較多,但方法學(xué)的比較研究很少,本文旨在比較精子DNA完整性常用檢測(cè)方法之間的差異,并分析DNA碎片與精子質(zhì)量的相關(guān)性。方法:選擇108例精液樣本,采用精子染色質(zhì)結(jié)構(gòu)分析試驗(yàn)(SCSA)和精子染色體擴(kuò)散實(shí)驗(yàn)(SCD)對(duì)樣本分別進(jìn)行精子DNA碎片檢測(cè)分析,對(duì)兩種方法的結(jié)果進(jìn)行比較,并與精液常規(guī)、精子形態(tài)以及患者年齡進(jìn)行相關(guān)性分析。結(jié)果:兩種方法檢測(cè)的精子DNA碎片化指數(shù)(DFI)存在顯著的一致性(P0.01),DFI與精子活動(dòng)率、前向運(yùn)動(dòng)精子百分率以及正常形態(tài)精子百分率呈顯著負(fù)相關(guān)(P0.01),與畸形精子指數(shù)呈顯著正相關(guān)(P_(SCSA)0.01,P_(SCD)0.05),SCSA得出的DFI與年齡呈顯著正相關(guān)(P0.01),而SCD法的DFI并未發(fā)現(xiàn)類(lèi)似的現(xiàn)象。結(jié)論:兩種方法檢測(cè)的結(jié)果對(duì)精子質(zhì)量均有較好的預(yù)測(cè)價(jià)值,精子DFI作為一個(gè)檢測(cè)指標(biāo),對(duì)男性生育力預(yù)測(cè)有一定的臨床價(jià)值。但不同方法之間的檢測(cè)結(jié)果差異較大,檢測(cè)方法的標(biāo)準(zhǔn)化有待進(jìn)一步研究。
[Abstract]:Objective: in recent years, sperm DNA fragments have attracted wide attention in predicting male fertility, and there are many methods for their detection, but there are few comparative studies on the methods. The aim of this study was to compare the differences of sperm DNA integrity test methods and to analyze the correlation between DNA fragments and sperm quality. Methods: one hundred and eight semen samples were selected. Sperm chromatin structure analysis test (SCSA) and sperm chromosome diffusion test (SCD) were used to detect and analyze sperm DNA fragments, and the results of the two methods were compared. The correlation analysis was made with semen routine, sperm morphology and patient's age. Results: there was significant consistency between the two methods in detecting sperm DNA fragmentation index (P 0.01). DFI was negatively correlated with sperm motility, forward motility and normal sperm morphology (P0.01). There was a significant positive correlation between DFI and age (P < 0.01). But the DFI of SCD method did not find similar phenomenon. Conclusion: the results of the two methods have a good predictive value for sperm quality, sperm DFI as a detection index. It has some clinical value to predict male fertility, but the results of different methods are quite different, and the standardization of test methods needs further study.
