外陰陰道假絲酵母菌病來源的熱帶假絲酵母菌多位點(diǎn)序列分型

【摘要】:目的:應(yīng)用多位點(diǎn)序列分型(MLST)分析38株熱帶假絲酵母菌的親緣關(guān)系,體外比較熱帶假絲酵母菌對(duì)常見抗真菌藥物的敏感性,檢測(cè)耐藥基因的表達(dá)水平。方法:通過對(duì)6對(duì)等位基因進(jìn)行測(cè)序,用e BURST分組和非加權(quán)配對(duì)算術(shù)平均法(UPGMA)聚類分析菌株的進(jìn)化和變異。微量肉湯稀釋法進(jìn)行體外抗真菌藥物敏感試驗(yàn),實(shí)時(shí)熒光定量聚合酶鏈反應(yīng)(q-RT-PCR)檢測(cè)耐藥基因CDR1、CDR2、ERG1、ERG3、ERG6、ERG9和ERG11表達(dá)。結(jié)果:38株菌株中獲得30個(gè)二倍體序列(DST),其中14個(gè)新DST,11個(gè)新等位基因,27個(gè)DST為單體型。e BURST分組,主要分成2組,大多數(shù)DST屬于單體型。聚類分析總共分18個(gè)進(jìn)化枝。阿尼芬凈、米卡芬凈、卡泊芬凈、氟康唑、伏立康唑、伊曲康唑、兩性霉素B、氟胞嘧啶、咪康唑、布康唑、克霉唑、特康唑、特比萘芬和制霉菌素14種抗真菌藥物的MIC90分別為0.030μg/ml、0.015μg/ml、0.500μg/ml、1.000μg/ml、0.125μg/ml、0.250μg/ml、1.000μg/ml、0.250μg/ml、2.000μg/ml、2.000μg/ml、0.250μg/ml、0.250μg/ml、256.000μg/ml和8.000μg/ml。絕大多數(shù)耐藥基因表達(dá)量較高的菌株可能在進(jìn)化枝11和14。結(jié)論:熱帶假絲酵母菌MLST變異較大,菌株之間遺傳距離較遠(yuǎn)。熱帶假絲酵母菌對(duì)特比萘芬普遍耐藥,對(duì)目前其他常用的抗真菌藥物多為敏感。
[Abstract]:Objective: to analyze the relationship of 38 strains of Candida tropicalis by multilocus sequence typing (MLST), and to compare the susceptibility of Candida tropicalis to common antifungal drugs in vitro. Methods: six pairs of alleles were sequenced. The evolution and variation of strains were analyzed by e BURST grouping and unweighted pairing arithmetic average method. In vitro antifungal susceptibility test was carried out by broth dilution method. Real-time fluorescence quantitative polymerase chain reaction (RT-PCR) was used to detect the resistance gene CDR1, CDR2ERG1, ERG3 and ERG6. ERG9 and ERG11 expression. Results 30 diploid sequences were obtained from 38 strains, of which 14 new DSTs and 11 new alleles were obtained. 27 DST were divided into haplotype. E BURST groups, and most of them were haplotypes. Cluster analysis consisted of 18 evolutionary branches. Fluconazole, Volconazole, itraconazole, amphotericin B, Fluocytosine, Miconazole, buconazole, clotrimazole, Teconazole. The MIC90 of terbinafine and nystatin were 0.030 渭 g / ml, 0.015 渭 g / ml and 0.500 渭 g / ml, respectively. 0.125 渭 g / ml 0.250 渭 g / ml 1.000 渭 g / ml 0.250 渭 g / ml 2.000 渭 g / ml 2.000 渭 g / ml. 0.250 渭 g / ml 0.250 渭 g / ml. 256.000 渭 g / ml and 8.000 渭 g / ml. Most strains with high expression of drug-resistant genes may be in Evolutionary branches 11 and 14. Conclusion:. The MLST variation of Candida tropicalis was significant. Candida tropicalis is generally resistant to terbinafine and is sensitive to other commonly used antifungal agents.
