【摘要】：目的:探討富血小板血漿(PRP)中血小板濃度對血小板聚集率的影響,初步確定誘聚劑二磷酸腺苷(ADP:5.0和15μmol/L)和花生四烯酸(AA:500.0μg/ml)對血小板濃度的允許范圍。方法:選取健康查體人群枸櫞酸鈉抗凝外周血標(biāo)本72例,每12例標(biāo)本混合為1組,分6組。將各組標(biāo)本混合后,分別制備不同水平的血小板濃度,利用海倫娜血小板聚集分析儀分別檢測AA和ADP誘導(dǎo)下的6組不同血小板濃度水平的血小板聚集率。結(jié)果:當(dāng)誘聚劑濃度升高,不同濃度的血小板聚集率會隨之增高。隨著血小板濃度升高,AA和ADP(15μmol/L)誘導(dǎo)的血小板聚集率均呈現(xiàn)先快速上升而后平緩的趨勢,其中當(dāng)Plt濃度低于200×10~9/L時,AA誘導(dǎo)的血小板聚集率隨著血小板濃度的降低出現(xiàn)陡降,而ADP誘導(dǎo)的血小板聚集率隨著血小板濃度的升高稍平緩些。當(dāng)Plt200×10~9/L,AA 500μg/ml和ADP 15μmol/L誘導(dǎo)時,不同濃度Plt的血小板聚集率變化平緩、波動較少,較穩(wěn)定。結(jié)論:進行血小板聚集率檢測時,AA和ADP誘聚劑對應(yīng)的適宜Plt濃度應(yīng)大于200×10~9/L。
[Abstract]:Objective: to investigate the effect of platelet concentration on platelet aggregation in platelet-rich plasma (PRP). ADP: 5.0 and 15 渭 mol / L) and AA: 500.0 渭 g / ml). Methods: 72 cases of peripheral blood samples with sodium citrate anticoagulant were selected. Every 12 samples were mixed into 1 group and divided into 6 groups. After each group was mixed, different levels of platelet concentration were prepared. The platelet aggregation rates of 6 groups induced by AA and ADP were measured by Helena platelet aggregation analyzer. Results: when the concentration of attractant increased. With the increase of platelet concentration, the platelet aggregation rate induced by AA and ADP(15 渭 mol / L increased rapidly first and then slowly. When the concentration of Plt was lower than 200 脳 10 ~ (9) / L, the platelet aggregation rate induced by ACA decreased sharply with the decrease of platelet concentration. However, the platelet aggregation rate induced by ADP was slightly slower with the increase of platelet concentration, when Plt200 脳 10 ~ (-1) / L was used. When AA 500 渭 g / ml and ADP 15 渭 mol/L were induced, the platelet aggregation rate of different concentrations of Plt changed slowly and fluctuated less. Conclusion: the suitable concentration of Plt corresponding to AA and ADP should be more than 200 脳 10 ~ (9) L ~ (-1) for platelet aggregation.
