血必凈通過(guò)Rho激酶信號(hào)通路對(duì)膿毒癥急性腎損傷后腎小管細(xì)胞的影響研究

【摘要】:目的探討早期應(yīng)用血必凈通過(guò)Rho激酶信號(hào)通路對(duì)膿毒癥急性腎損傷后腎小管細(xì)胞凋亡的保護(hù)作用。方法 2014年8月—2015年6月,54只雄性SD大鼠采用隨機(jī)數(shù)字表法分為假手術(shù)組、膿毒癥組、血必凈組,每組18只。膿毒癥組和血必凈組以盲腸結(jié)扎穿孔(CLP)法制造膿毒癥模型,膿毒癥組大鼠CLP后0.5 h尾靜脈注射0.9%氯化鈉溶液4 ml/kg;血必凈組大鼠CLP后0.5 h尾靜脈注射血必凈注射液4 ml/kg。造模后6、24 h每組分別選取9只大鼠采用激光多普勒血流儀測(cè)定血流灌注(BPU)情況,采用全自動(dòng)生化儀檢測(cè)血肌酐、尿素氮、肌酐清除率(Cr Cl),病理組織學(xué)檢測(cè)評(píng)定腎小管損傷評(píng)分,采用原位末端標(biāo)記(TUNEL)法計(jì)算凋亡指數(shù)(AI),Western blotting法檢測(cè)Rho相關(guān)卷曲螺旋形成蛋白激酶1(ROCK-1)、肌球蛋白磷酸酶靶亞基1(MYPT-1)、含半胱氨酸的天冬氨酸蛋白水解酶3(Caspase-3)的表達(dá)水平。結(jié)果 3組大鼠造模后6 h BPU及造模后24 h BPU、血肌酐、尿素氮、Cr Cl比較,差異均有統(tǒng)計(jì)學(xué)意義(P0.05);其中造模后6 h,膿毒癥組大鼠BPU較假手術(shù)組升高,血必凈組大鼠BPU較假手術(shù)組升高、較膿毒癥組降低(P0.05);造模后24 h,膿毒癥組大鼠BPU較假手術(shù)組降低,膿毒癥組大鼠血肌酐、尿素氮、Cr Cl較假手術(shù)組升高,血必凈組大鼠尿素氮、Cr Cl較假手術(shù)組升高,血必凈組大鼠BPU較膿毒癥組升高,血必凈組大鼠血肌酐、尿素氮、Cr Cl較膿毒癥組降低(P0.05)。造模后6 h,3組腎小管損傷評(píng)分、AI比較,差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);造模后24 h,3組腎小管損傷評(píng)分、AI比較,差異有統(tǒng)計(jì)學(xué)意義(P0.05);其中膿毒癥組腎小管損傷評(píng)分、AI較假手術(shù)組升高,血必凈組腎小管損傷評(píng)分、AI較假手術(shù)組升高,血必凈組腎小管損傷評(píng)分、AI較膿毒癥組降低(P0.05)。造模后6、24 h,3組ROCK-1、MYPT-1和Caspase-3表達(dá)水平比較,差異均有統(tǒng)計(jì)學(xué)意義(P0.05);其中造模后6 h,膿毒癥組和血必凈組ROCK-1、MYPT-1和Caspase-3表達(dá)水平較假手術(shù)組升高(P0.05);造模后24 h,膿毒癥組ROCK-1、MYPT-1和Caspase-3表達(dá)水平較假手術(shù)組升高,血必凈組ROCK-1、MYPT-1表達(dá)水平較假手術(shù)組升高,血必凈組ROCK-1、MYPT-1和Caspase-3表達(dá)水平較膿毒癥組降低(P0.05)。結(jié)論早期應(yīng)用血必凈可以通過(guò)Rho激酶信號(hào)通路減少腎小管細(xì)胞在膿毒癥進(jìn)程中的凋亡,改善病理學(xué)改變,減輕膿毒癥導(dǎo)致的急性腎損傷。
[Abstract]:Objective to investigate the early application of Xuebijing via Rho kinase signal pathway on apoptosis of renal tubular cells in sepsis after acute kidney injury protection. Methods from August 2014 to June 2015, 54 male SD rats were randomly divided into sham operation group, sepsis group and Xuebijing group, 18 rats in each group.
