NaOH蝕刻玻碳電極的大腸桿菌DNA電化學(xué)生物傳感器的構(gòu)建及檢測

【摘要】:以NaOH蝕刻后的玻碳電極為基底制備了高靈敏與高選擇性的大腸桿菌電化學(xué)DNA傳感器。經(jīng)NaOH蝕刻后的玻碳電極被活化且在電極表面形成羧基層,為DNA探針的固定提供了更多位點(diǎn),在偶聯(lián)劑EDC/NHS的作用下,端氨修飾的探針DNA與羧基的羧氨反應(yīng)使其以肽鍵的形式固定在電極表面,極大地提升了傳感器的靈敏度。通過電化學(xué)循環(huán)伏安法(CV)和差分脈沖法(DPV)對所制備傳感器的靈敏度和選擇性進(jìn)行表征,得到該傳感器對大腸桿菌的線性檢測范圍為12.5~62.5 nmol/L,檢出限可達(dá)1.20×10~(-9)mol/L。
[Abstract]:A highly sensitive and selective E. coli electrochemical DNA sensor was prepared on the substrate of NaOH etched glassy carbon electrode. The glassy carbon electrode etched by NaOH is activated and forms a carboxy base on the electrode surface, which provides more sites for the fixation of DNA probe. Under the action of coupling agent EDC/NHS, The reaction of ammonia-terminated probe DNA with carboxy group makes it fixed on the electrode surface in the form of peptide bond, which greatly improves the sensitivity of the sensor. The sensitivity and selectivity of the sensor were characterized by electrochemical cyclic Voltammetric method (CV) and differential pulse method (DPV).
