金仿生礦化復(fù)合酶的制備及其初步的醫(yī)學(xué)與環(huán)境應(yīng)用

【摘要】:貴金屬納米簇尤其金納米簇(Au NCs)以其優(yōu)良的尺寸效應(yīng)和光電功能優(yōu)勢(shì),被廣泛應(yīng)用于生物標(biāo)記、酶催化、成像分析、以及傳感檢測(cè)等領(lǐng)域。本文采用生物仿生礦化途徑,一方面制備了一種金礦化的葡萄糖氧化酶(GOx)復(fù)合物,用以修飾核酸探針并結(jié)合磁分離技術(shù),發(fā)展了一種電化學(xué)信號(hào)放大的夾心法,用于炭疽病毒DNA序列的超靈敏檢測(cè)及其單堿基突變分析;另一方面,開發(fā)了一種血紅素(Hemin)介導(dǎo)金礦化的多功能模擬酶,用以標(biāo)記甲胎蛋白(AFP)抗體,并借助其具有的過氧化物酶催化與金催化銀沉積功能,實(shí)現(xiàn)了對(duì)人血清中AFP的快速、靈敏、可視化檢測(cè);與此同時(shí),將該模擬酶與Fe3O4負(fù)載的三聚氰胺甲醛樹脂(m PMF)介孔聚合物相結(jié)合,開發(fā)了一種可回收利用的多功能復(fù)合材料,用于環(huán)境有機(jī)毒物(染料)的高效吸附與催化降解。主要內(nèi)容包括:(1)建立一種基于金仿生礦化GOx的電化學(xué)傳感技術(shù),應(yīng)用于炭疽病毒DNA序列的檢測(cè)及其單堿基突變分析(第二章)。一方面,以GOx蛋白為還原劑和穩(wěn)定劑,通過原位金仿生礦化途徑制得GOx-Au復(fù)合酶,用以標(biāo)記經(jīng)酪胺修飾的核酸檢測(cè)探針,同時(shí),以磁性顆粒負(fù)載經(jīng)酪胺修飾的核酸捕獲探針,進(jìn)而將二者與目標(biāo)鏈雜交形成“三明治”型核酸復(fù)合物;另一方面,借助GOx-Au復(fù)合酶中GOx催化葡萄糖分解得到雙氧水,進(jìn)而利用其中礦化金具有的類過氧化物酶功能催化雙氧水氧化酪胺偶聯(lián),籍以連接DNA雜交反應(yīng)后的捕獲探針和檢測(cè)探針,并借助金催化金沉積途徑放大其電化學(xué)信號(hào),建立了一種電化學(xué)傳感新技術(shù),實(shí)現(xiàn)了對(duì)血液中炭疽病毒DNA的超靈敏檢測(cè)及其單堿基突變分析。由此建立的基于金仿生礦化GOx催化信號(hào)放大途徑的電化學(xué)傳感方法,突破了低含量短鏈DNA的檢測(cè)瓶頸,可望為臨床炭疽病毒感染的早期診斷提供一種新的檢測(cè)工具。(2)基于血紅素介導(dǎo)金礦化途徑的模擬酶復(fù)合物的制備及其用于AFP的比色分析(第三章)。采用生物礦化途徑,以Hemin為模板于堿性條件下介導(dǎo)金礦化,制得Hemin-Au模擬酶復(fù)合物。研究表明,Hemin-Au兼具Hemin類過氧化物酶和金的雙重催化功能。以肝癌標(biāo)志物AFP的免疫分析為例,將該模擬酶復(fù)合物標(biāo)記AFP抗體,并結(jié)合酶聯(lián)免疫吸附試驗(yàn)途徑,建立了酶催化底物顯色和金催化銀沉積反應(yīng)兩種信號(hào)放大途徑,用以分別實(shí)現(xiàn)了對(duì)血液中AFP的高靈敏比色檢測(cè)。(3)基于磁性顆粒負(fù)載m PMF介孔聚合物的模擬酶復(fù)合材料的制備及其用于環(huán)境染料的吸附與催化降解(第四章)。采用水熱法合成磁性納米顆粒,進(jìn)而借助三聚氰胺和甲醛的羥基化縮聚反應(yīng)形成m PMF介孔聚合物,并包覆于磁性納米顆粒表面,制得Fe3O4@m PMF磁性復(fù)合物。然后,通過化學(xué)鍵合方法將Hemin-Au模擬酶負(fù)載于Fe3O4@m PMF顆粒上,制得具有高比表面積、強(qiáng)催化功能、可回收利用的多功能復(fù)合材料。以亞甲基藍(lán)(MB)和羅丹明B(RhB)為例,系統(tǒng)地考察了該復(fù)合材料吸附與催化降解常見環(huán)境染料的性能。結(jié)果表明,這種基于mPMF介孔聚合物的磁性模擬酶復(fù)合材料,可望作為一種綠色、高效的吸附與催化新材料,廣泛應(yīng)用于染料等環(huán)境毒物的高效處理。
