酵母菌油脂含量測定方法的比較研究

【摘要】:為確定一種快速簡便、準(zhǔn)確可靠的酵母菌油脂含量測定方法,對磷酸香草醛顯色法、氧化酶法與重量法進(jìn)行比較分析。結(jié)果表明:在測定酵母菌體總脂含量時(shí)重量法準(zhǔn)確度和精確度高,成本低,但操作過程煩瑣、耗時(shí)長,溶劑有毒易污染環(huán)境;磷酸香草醛顯色法的平均回收率約為93%,準(zhǔn)確度和精確度較高,耗時(shí)少,最低檢測限約為0.08 g/L,測定時(shí)需與相應(yīng)的菌體油脂標(biāo)準(zhǔn)曲線匹配使用,其測定發(fā)酵液油脂含量與重量法差異不顯著(P0.05),可以作為酵母菌油脂含量測定的方法;在測定酵母菌體內(nèi)甘油三酯含量時(shí),氧化酶法平均回收率達(dá)到98.0%,準(zhǔn)確度和精確度高,該方法操作簡便,耗時(shí)少,最低檢測限為1.2 g/L;氧化酶法需與甘油三酯標(biāo)準(zhǔn)曲線匹配使用,同時(shí)還需對細(xì)胞進(jìn)行破壁,但由于其快速準(zhǔn)確性,也不失為一種酵母菌油脂含量測定的好方法。
[Abstract]:In order to determine a rapid, simple, accurate and reliable method for the determination of yeast oil, the colorimetric method of vanillin phosphate, oxidase method and gravimetric method were compared and analyzed. The results showed that the gravimetric method had high accuracy and accuracy and low cost in the determination of total lipid content of yeast, but the operation process was cumbersome, the time was long, and the solvent was toxic and easy to pollute the environment. The average recovery of vanillin phosphate colorimetric method is about 933, the accuracy and accuracy are higher, the time consuming is less, the minimum detection limit is about 0.08 g / L, and the determination needs to be matched with the corresponding bacterial oil standard curve. The determination of lipid content in fermentation broth was not significantly different from that by gravimetric method, which could be used as a method for the determination of lipid content of yeast, and the average recovery rate of oxidase method was 98.0 and the accuracy and accuracy were high when determining the content of triglyceride in yeast. The method is simple and time-consuming, with a minimum detection limit of 1.2 g / L. Oxidase method is used to match the standard curve of triglyceride and to break the cell wall, but because of its rapid accuracy, It is also a good method for the determination of the oil content of Saccharomyces cerevisiae.
