新型采油工程菌的構(gòu)建與性能評價

【摘要】:隨著我國經(jīng)濟(jì)的快速發(fā)展,對石油的需求量與日俱增。微生物采油技術(shù)具有成本低和不污染環(huán)境等優(yōu)點,受到廣泛關(guān)注,但目前學(xué)者的目光主要集中于對單一菌株采油效果或幾種菌株復(fù)合采油效果的研究,而將基因工程方法應(yīng)用于微生物采油則鮮見報道,因此有必要在這方面展開深入的研究。本文首先從產(chǎn)生物表面活性劑的大腸桿菌中提取出4種鼠李糖脂表達(dá)質(zhì)粒,應(yīng)用電擊轉(zhuǎn)化的方法將鼠李糖脂表達(dá)質(zhì)粒轉(zhuǎn)入產(chǎn)生物聚合物的菌株內(nèi),構(gòu)建出3株能同時產(chǎn)生生物表面活性劑和生物聚合物的轉(zhuǎn)化菌株,之后通過評價各轉(zhuǎn)化菌株的產(chǎn)物性能,優(yōu)選出一株性能最好的菌株WS2作為新型采油工程菌。然后對該菌株的適應(yīng)性進(jìn)行評價,實驗結(jié)果表明,該菌株溫度適應(yīng)范圍為35~75℃,pH值適應(yīng)范圍為5~10,穩(wěn)定時間在40d以內(nèi),同時還具有良好的鹽度適應(yīng)性和抗剪切性能。最后對菌株WS2的驅(qū)油效果進(jìn)行評價,實驗結(jié)果表明,新型菌株WS2的驅(qū)油效果好于產(chǎn)生物聚合物菌株SP1和產(chǎn)生物表面活性劑菌株BS1,水驅(qū)后采收率可提高16.66%,分別比菌株SP1和BS1高出3.32%和6.43%。研究結(jié)果為微生物采油技術(shù)的推廣和應(yīng)用提供了理論依據(jù)與技術(shù)支持。
[Abstract]:With the rapid development of economy in China, the demand for oil is increasing day by day. Microbial oil recovery technology has the advantages of low cost and no pollution to the environment. At present, however, the focus of scholars is mainly on the oil recovery effect of a single strain or several strains, but the application of genetic engineering to microbial oil recovery is rarely reported. Therefore, it is necessary to carry out in-depth research in this field. Firstly, four kinds of rhamnolipid expression plasmids were extracted from Escherichia coli, which produced surfactant. The expression plasmid of rhamnolipid was transferred into the polymer producing strain by electroporation method. Three transformed strains were constructed which could produce biosurfactant and biopolymer simultaneously, and then evaluated the product properties of each transformed strain. A strain WS2 with the best performance was selected as a new oil recovery engineering strain. The adaptability of the strain was evaluated. The experimental results showed that the temperature adaptation range of the strain was 35 ~ 75 鈩,
