酸性土壤中接種耐酸根瘤菌對(duì)豆科植物根際微生態(tài)的影響

【摘要】:【目的】接種耐酸豆科根瘤菌可以提高豆科植物耐酸能力。應(yīng)用PLFA等方法研究接種耐酸根瘤菌對(duì)根際土壤微生態(tài)的綜合影響,從土壤角度闡明接種耐酸根瘤菌緩解土壤酸度對(duì)豆科植物脅迫的機(jī)制。【方法】盆栽條件下分別向種植于p H 4.8酸性土壤中的葛藤和苜蓿植株相應(yīng)接種耐酸葛藤根瘤菌068、389、390與耐酸苜蓿根瘤菌91512、91522、91532,于接種后30 d、60 d、90 d、120 d、150 d取樣,分析耐酸根瘤菌對(duì)土壤養(yǎng)分、可培養(yǎng)微生物、微生物群落結(jié)構(gòu)多樣性的影響!窘Y(jié)果】1)接種耐酸葛藤和苜蓿根瘤菌處理后,根際土壤p H由4.8顯著提高至6.0左右,有效磷、速效鉀、銨態(tài)氮、硝態(tài)氮及有機(jī)質(zhì)含量的增加效果顯著(P0.05),至120 d達(dá)到最高。120 d樣品的有效磷、銨態(tài)氮、硝態(tài)氮及有機(jī)質(zhì)含量分別為23.16~48.68 mg/kg、61.21~81.96mg/kg、65.05~86.38 mg/kg和11.85~12.87 g/kg,分別比未接種對(duì)照提高24.8%~162.4%、16.6%~56.2%、145.4%~225.8%、1.4%~10.1%;2)接種苜蓿和葛藤耐酸根瘤菌后土壤中可培養(yǎng)微生物數(shù)量顯著提高(P0.05),土壤中可培養(yǎng)細(xì)菌、真菌和放線菌數(shù)量在120 d達(dá)到峰值,比同期未接種對(duì)照分別提高了61.5%~348.4%、3.4%~441.7%和18.9%~255.2%,分別達(dá)到48.00×10~2~133.3×10~7、20.11×10~4~155.9×10~4和3.21×10~4~9.59×10~4 cfu/g鮮土;3)PLFA分析表明,雖然接種耐酸葛藤、苜蓿根瘤菌處理的根際土壤的特征脂肪酸種類數(shù)與未接種處理相比并無(wú)明顯差異,但接種葛藤或苜蓿根瘤菌均明顯降低了根際土壤中異構(gòu)PLFA/反異構(gòu)PLFA值,顯示土壤根際微生態(tài)的穩(wěn)定性得到提高。【結(jié)論】在種植豆科植物的酸性土壤中接種相應(yīng)耐酸根瘤菌能顯著提高根際土壤p H,提高根際土壤有機(jī)質(zhì)含量和速效氮磷養(yǎng)分含量,緩解土壤酸性對(duì)根際微生物的脅迫。
[Abstract]:Objective to improve the acid tolerance of leguminous plants by inoculating acid-tolerant leguminous Rhizobium. PLFA and other methods were used to study the comprehensive effect of inoculating acid-tolerant Rhizobium on the microecology of rhizosphere soil, and to elucidate the mechanism of inoculating acid-tolerant Rhizobium to alleviate the stress of leguminous plants by soil acidity. [methods] under pot culture conditions, the acid-tolerant Gegeraria and Medicago sativa plants planted in pH 4.8 acid soil were inoculated with acid-resistant Gegeraria lobata 068389390 and acid-tolerant Medicago sativa 91512, 91522, 91532, respectively, 30 days, 60 days, 90 days, 120 days, 30 days, 60 days, 90 days, 120 days, 90 days and 120 days, respectively. The effects of acid-tolerant Rhizobium on soil nutrients, culturable microorganisms and microbial community structure diversity were analyzed by 150 d sampling. [results] 1) after inoculating with acid-tolerant Pueraria and Medicago sativa Rhizobium, the pH of rhizosphere soil increased significantly from 4.8 to 6.0, and the contents of available phosphorus, available potassium, ammonium nitrogen, nitrate nitrogen and organic matter increased significantly (P 0.05).
