煙蚜線粒體基因組全序列及系統(tǒng)發(fā)育分析(英文)

【摘要】:【目的】蚜蟲是雜食性農(nóng)業(yè)害蟲。本研究旨在通過線粒體基因組分析更好地了解蚜科昆蟲的系統(tǒng)發(fā)育關(guān)系!痉椒ā拷Y(jié)合第二代測序和PCR擴(kuò)增技術(shù)獲得了煙蚜Myzus persicae線粒體基因組全序列,與蚜科其他昆蟲進(jìn)行了對(duì)比分析;以貝葉斯法和最大似然法基于13個(gè)蛋白編碼基因?qū)ρ量七M(jìn)行了系統(tǒng)發(fā)育分析。【結(jié)果】煙蚜線粒體基因組(Gen Bank登錄號(hào):KU_236024)序列全長17 832 bp,A+T含量84.1%,AT偏斜為0.094,GC偏斜為-0.296。包含13個(gè)蛋白編碼基因(cox1-3,nad1-6,nad4L,atp6,atp8和cytb),22個(gè)tRNA,2個(gè)rRNA基因(rrn L和rrn S)和2個(gè)長的非編碼區(qū),其基因排列順序與已知的蚜科昆蟲相似,除了nad4以單獨(dú)的T結(jié)尾,所有的蛋白編碼基因均以ATN作為起始密碼子,TAA作為終止密碼子。在煙蚜線粒體基因組中,tRNAGlu和tRNAPhe中間有一段307 bp的非編碼區(qū),該編碼區(qū)包含2個(gè)重復(fù)單元,煙蚜的控制區(qū)長2 531 bp,是所有測序蚜蟲線粒體基因組中最長的。rrn L的二級(jí)結(jié)構(gòu)包含6個(gè)結(jié)構(gòu)域,44個(gè)莖環(huán)結(jié)構(gòu);rrn S的二級(jí)結(jié)構(gòu)有3個(gè)結(jié)構(gòu)域,24個(gè)莖環(huán)結(jié)構(gòu);跓煼凼推渌20種昆蟲的13個(gè)蛋白編碼基因重建的BI和ML系統(tǒng)發(fā)育樹,與傳統(tǒng)形態(tài)學(xué)分類結(jié)果一致!窘Y(jié)論】蚜亞科和長管蚜亞科的單系性得到了很好的支持;在長管蚜亞科的分支中,M.persicae與D.noxia聚成一支,并且C.salicicola位于進(jìn)化枝的底部。本研究結(jié)果為蚜科系統(tǒng)發(fā)生關(guān)系重建積累了有價(jià)值的數(shù)據(jù)資料。
[Abstract]:[objective] aphids are omnivorous agricultural pests. The aim of this study was to better understand the phylogenetic relationship of aphids by mitochondrial genome analysis. [methods] complete mitochondrial genome sequences of aphid Myzus persicae were obtained by using the second generation sequencing and PCR amplification techniques. The results were compared with other insects of aphids. Phylogenetic analysis of aphids was carried out by using Bayesian method and maximum likelihood method based on 13 protein coding genes. [results] the total length of mitochondrial (Gen Bank accession number: KU_236024) sequence of APHIS fumigatus was 17832 bp,A T content 84.1%. The deviation of AT and GC were 0.094 and-0.296 respectively. It contains 13 protein-coding genes (cox1-3,nad1-6,nad4L,atp6,atp8 and cytb), 22 tRNA,2, (rrn L and rrn S), and 2 long non-coding regions), the sequence of which is similar to that of known aphids. Except that nad4 ends with a single T, all the protein coding genes use ATN as the starting codon and TAA as the terminating codon. In the mitochondrial genome of APHIS fumigatus, there is a non-coding region of 307bp between tRNAGlu and tRNAPhe. The coding region contains two repeat units, and the control region of APHIS fumigatus is 2531 bp,. The secondary structure of rrn L contains 6 domains and 44 stem rings. The secondary structure of rrn S has 3 domains and 24 stem rings. The phylogenetic tree of BI and ML based on 13 protein coding genes of Bemisia Tabaci and other 20 species of insects is consistent with the results of traditional morphological classification. [conclusion] the monophyletic characters of aphid subfamily and long tube aphid subfamily are well supported. In the branches of the aphids, M.persicae and D.noxia form one branch, and C.salicicola is located at the bottom of the evolutionary branch. The results of this study have accumulated valuable data for the phylogenetic relationship reconstruction of aphids.
