黑曲霉YF1菌株對(duì)煙嘧磺隆的降解條件研究

【摘要】:黑曲霉(Aspergillus niger)YF1菌株是本實(shí)驗(yàn)室篩選得到的對(duì)煙嘧磺隆具有較強(qiáng)降解能力的菌株。為優(yōu)化YF1菌株的降解條件,本試驗(yàn)分別對(duì)葡萄糖濃度、培養(yǎng)溫度、初始pH和轉(zhuǎn)速進(jìn)行單因素試驗(yàn),結(jié)果表明,YF1菌株降解煙嘧磺隆的最適條件為:葡萄糖濃度5g/L、培養(yǎng)溫度30℃、初始pH值6.50和轉(zhuǎn)速50r/min。本試驗(yàn)在研究其降解條件的同時(shí),發(fā)現(xiàn)溶液的pH均出現(xiàn)下降,隨后采用高效液相色譜檢測(cè)草酸、L-蘋果酸、α-酮戊二酸、乙酸、檸檬酸、D-蘋果酸、琥珀酸的標(biāo)準(zhǔn)品,通過(guò)保留時(shí)間確定樣品中的有機(jī)酸為草酸,但未檢測(cè)出其它6種有機(jī)酸,因此,可以確定YF1菌株可以代謝葡萄糖產(chǎn)生草酸。本試驗(yàn)初步明確了黑曲霉YF1菌株對(duì)煙嘧磺隆的降解特性,為該菌株的應(yīng)用奠定了基礎(chǔ)。
