氨氧化菌的分離鑒定及其在羅氏沼蝦育苗廢水中的應用

【摘要】:氨氮轉(zhuǎn)化細菌在養(yǎng)殖水生態(tài)系統(tǒng)的物質(zhì)循環(huán)和水體凈化的過程中起著重要作用。該研究從養(yǎng)殖池塘底泥中篩選具有氨氮去除能力的細菌,通過生化和分子生物學技術(shù)進行細菌鑒定,并用于羅氏沼蝦育苗用水的氨氮凈化。結(jié)果表明:分離獲得的氨氮去除菌YX0703能在無機氮培養(yǎng)基和有機氮培養(yǎng)基中生長,既具有氨氮氧化活性,也能將NO_3~-轉(zhuǎn)化為NO_2~-,再將NO_2~-轉(zhuǎn)化為N_2,經(jīng)鑒定,該菌為人蒼白桿菌(Ochrobactrum anthropi);該菌株攜帶亞硝酸還原酶基因(Nitrite reductase K gene,nirK),藥物最低抑菌濃度檢測發(fā)現(xiàn),其對頭孢類抗菌素耐藥,對喹諾酮類、美羅培南和四環(huán)素等抗菌藥物敏感;搖床轉(zhuǎn)速150 r·min~(-1)時,在25、30℃條件下,菌株YX0703對羅氏沼蝦育苗廢水的氨氮去除率分別為96.98%和97.44%。綜上,分離獲得的氨氮去除菌YX0703是人蒼白桿菌,其既能利用無機氮又能利用有機氮,既具有氨氧化活性又有反硝化活性,可用于羅氏沼蝦育苗廢水的氨氮凈化。
[Abstract]:Ammonia-nitrogen conversion bacteria play an important role in the process of material circulation and water purification in aquaculture water ecosystem. In this study, the bacteria with ammonia nitrogen removal ability were screened from the sediment of cultured ponds. The bacteria were identified by biochemical and molecular biological techniques, and were used to purify ammonia nitrogen in the water used for the breeding of Macrobrachium rosenbergii (Macrobrachium rosenbergii). The results showed that the ammonia nitrogen removal bacteria YX0703 could grow in inorganic nitrogen medium and organic nitrogen medium, which not only had ammonia nitrogen oxidation activity, but also transformed NO_3~- into NO_2~-, and then transformed NO_2~- into N\ + 2\ +\ {2\}. The results showed that the bacteria could grow in inorganic nitrogen medium and organic nitrogen medium. The bacterium is called (Ochrobactrum anthropi);. The strain carried the nitrite reductase gene (Nitrite reductase K gene,nirK). The minimal inhibitory concentration (MIC) of the drug showed that it was resistant to cephalosporins and sensitive to quinolones, meropenem and tetracycline, and was sensitive to antibiotics such as quinolones, meropenem and tetracycline.
