貂糞堆肥微生物復(fù)合菌劑的制備及效果評(píng)價(jià)

【摘要】:隨著經(jīng)濟(jì)和社會(huì)的迅速發(fā)展,我國(guó)的養(yǎng)殖業(yè)趨于規(guī)模化和集約化,隨之帶來了嚴(yán)重的養(yǎng)殖業(yè)污染問題。堆肥能夠?qū)崿F(xiàn)養(yǎng)殖業(yè)廢棄物的無害化和資源化處理,但是傳統(tǒng)的堆肥存在耗時(shí)長(zhǎng)、發(fā)酵不徹底等諸多弊端,因此在堆肥中添加微生物復(fù)合菌劑以實(shí)現(xiàn)養(yǎng)殖業(yè)廢棄物的快速堆肥處理具有重要的現(xiàn)實(shí)意義。本論文旨在制備用于貂糞堆肥的微生物復(fù)合菌劑以實(shí)現(xiàn)貂糞的快速堆肥無害化處理。具體的研究?jī)?nèi)容如下:(1)建立普通好氧堆肥,在堆肥過程中的不同階段取樣,從樣品中篩選到堆肥微生物共20余種,經(jīng)過菌落形態(tài)特征方面的比較進(jìn)行分類合并,最后得到Al,A2,A3,A4, A5, A6, A7, A8共8種堆肥微生物;(2)對(duì)篩選的堆肥微生物菌種進(jìn)行16S rDNA鑒定以及其他生理生化功能鑒定,最后確定分別與Bacillus amyloliquefaciens, Bacillus flexus、Bacillus subtilis, Ureibacillus thermosphaericus, Geobacillus sp., Geobacillus pallidus, Bacillus licheniformis, Ureibacillus sp.具有很高的相似性,并且都具有各自獨(dú)特的功能,將其擴(kuò)大培養(yǎng)后輔以載體制成微生物復(fù)合菌劑ABTNL Ⅱ備用;(3)將自主研發(fā)的微生物復(fù)合菌劑接種于貂糞堆肥中,檢測(cè)物理、化學(xué)以及生物學(xué)指標(biāo)并進(jìn)行比較,結(jié)果顯示,實(shí)驗(yàn)組當(dāng)天即進(jìn)入高溫期(55℃以上),之后持續(xù)14d,最高溫度達(dá)到64.1℃,顯著高于對(duì)照組的59℃;而第3d才開始升溫的對(duì)照組的高溫期僅僅持續(xù)了7d;實(shí)驗(yàn)組第5d已基本上沒有臭味,但對(duì)照組第7d仍能聞到臭味;貂糞堆肥過程中,添加微生物復(fù)合菌劑的實(shí)驗(yàn)組的各項(xiàng)化學(xué)指標(biāo)(pH值、C/N、電導(dǎo)率、E4/E6、水溶性氨氮和硝氮)的變化與空白對(duì)照組相比均更為明顯且更加理想;實(shí)驗(yàn)組的發(fā)芽指數(shù)在第10d即達(dá)到80%以上,堆肥結(jié)束后達(dá)95%以上,而對(duì)照組在堆肥結(jié)束后仍未能達(dá)到80%。綜上所述,貂糞堆肥中添加微生物復(fù)合菌劑不僅能夠促進(jìn)其快速升溫,而且能夠大大延長(zhǎng)高溫持續(xù)時(shí)間,縮短堆肥周期,加速堆肥中大分子營(yíng)養(yǎng)物質(zhì)和植物毒性物質(zhì)的降解和腐殖化。可以將腐熟產(chǎn)品制成有機(jī)肥或生物有機(jī)肥,從而有效地保證了貂糞的無害化處理和資源化利用。
[Abstract]:With the rapid development of economy and society, China's aquaculture industry tends to scale and intensive, resulting in serious pollution problems. Compost can realize harmless and resource treatment of aquaculture waste, but traditional compost has many disadvantages such as time-consuming, fermentation incomplete and so on. Therefore, it is of great practical significance to add microbial compound fungicides into compost to realize rapid composting treatment of aquaculture waste. The aim of this paper is to prepare microbial composting agent for mink manure composting to realize the harmless treatment of mink manure composting. The specific research contents are as follows: (1) establishing common aerobic compost, sampling in different stages of composting process, screening out more than 20 kinds of compost microorganism from the sample, and combining the colony morphological characteristics. Finally, eight composting microorganisms, Al,A2,A3,A4, A5, A6, A7 and A8, were obtained. (2) 16s rDNA and other physiological and biochemical functions were used to identify the selected microbial strains of compost. Finally, the strain was identified with Bacillus amyloliquefaciens, Bacillus flexus,Bacillus subtilis, Ureibacillus thermosphaericus, Geobacillus sp., Geobacillus pallidus, Bacillus licheniformis, Ureibacillus sp., respectively.
