【摘要】：以檸條根際土壤為試材,采用CAS(鉻天青S)選擇性培養(yǎng)基分離篩選具有產(chǎn)鐵載體功能的根際促生菌株,通過16S r DNA序列分析進(jìn)行菌株鑒定,并測(cè)定菌株解磷、固氮、產(chǎn)IAA(吲哚乙酸)和ACC(1-氨基環(huán)丙烷-1-羧酸)脫氨酶等促生活性,通過盆栽試驗(yàn)分析驗(yàn)證菌株Fe11對(duì)檸條幼苗生長的影響。結(jié)果表明:共分離出14株產(chǎn)鐵載體根際促生菌,其中6株菌具備較強(qiáng)的產(chǎn)鐵載體能力。所有菌株的16S r DNA序列同源性與假單胞菌達(dá)到99%~100%,假單胞菌屬細(xì)菌是檸條根際產(chǎn)鐵載體的優(yōu)勢(shì)菌群。多數(shù)菌株兼具2種以上促生活性,菌株Fe9、Fe11和Fe13表現(xiàn)出多種促生潛能。回接試驗(yàn)證實(shí),接種菌株Fe11使檸條幼苗的株高、根長、葉片數(shù)、鮮質(zhì)量和干質(zhì)量分別增長17.25%、6.68%、17.75%、42.51%和20.67%,表明菌株Fell對(duì)檸條的生長具有良好的促進(jìn)效果。
[Abstract]:The rhizosphere soil of Caragana korshinskii was used to isolate and screen the rhizosphere growth-promoting strains with the function of iron producing carrier by CAS (Chromium-azurol S) selective medium. The strains were identified by 16s r DNA sequence analysis, and the phosphorus and nitrogen fixation were determined. IAA (indoleacetic acid) and ACC (1-aminocyclopropane-1-carboxylic acid) deaminase were produced. The effects of strain Fe11 on the growth of Caragana korshinskii seedlings were tested by pot experiment. The results showed that 14 strains of iron-producing carriers were isolated and 6 of them had strong ferric carrier capacity. The homology of 16s r DNA sequence between all strains and Pseudomonas was 990.The genus Pseudomonas was the dominant carrier for iron production in the rhizosphere of Caragana korshinskii. Most strains had two or more probiotic activities, and Fe9,Fe11 and Fe13 showed a variety of probiotic potential. The results showed that the plant height, root length, leaf number, fresh weight and dry mass of Caragana seedlings increased 17.25%, 6.68%, 17.75% and 20.67%, respectively, by inoculating strain Fe11. The results showed that the strain Fell had a good promoting effect on the growth of Caragana korshinskii.
