Act0988拮抗菌株鑒定及培養(yǎng)條件與菌株生長(zhǎng)關(guān)系研究

【摘要】:Act0988菌株產(chǎn)對(duì)柑橘青霉等多種霉菌具抗菌活性的代謝產(chǎn)物,對(duì)菌株進(jìn)行鑒定、研究培養(yǎng)條件與菌株生長(zhǎng)的關(guān)系,以為后續(xù)開發(fā)菌株產(chǎn)抗菌物質(zhì)的培養(yǎng)基篩選及發(fā)酵工藝技術(shù)的研究奠定基礎(chǔ)。以菌株的形態(tài)、培養(yǎng)特征、生理生化特征及16S rDNA序列分析進(jìn)行菌種鑒定,三角瓶液體振蕩培養(yǎng)研究碳氮源、溫度、pH及氧與菌株生長(zhǎng)的關(guān)系。結(jié)果表明,Act0988菌株為多產(chǎn)色鏈霉菌(Streptomyces polychromogenes),菌株生長(zhǎng)最佳碳源為可溶性淀粉,發(fā)酵液生物量5.8 mg/mL;酵母膏與蛋白胨為最佳氮源,生物量5.7 mg/mL左右。菌株最適溫度28℃,生物量6.0 mg/mL。最適初始pH7.0,生物量6.2 mg/mL;最適裝液量60 mL/500mL,生物量6.3 mg/mL。Act0988菌株易培養(yǎng),發(fā)酵液抗菌活性強(qiáng),菌株產(chǎn)抗菌物質(zhì)具有突出的開發(fā)利用潛力。
[Abstract]:Act0988 strains produced metabolites with antibacterial activity against many fungi, such as Penicillium Citrus. The strains were identified, and the relationship between the culture conditions and the growth of the strains was studied. It lays a foundation for the further development of the culture medium for the production of antimicrobial substances and the study of fermentation technology. The strain morphology, culture characteristics, physiological and biochemical characteristics and 16s rDNA sequence analysis were used to identify the strain. The relationship between carbon and nitrogen source, temperature, pH and oxygen and the growth of the strain was studied by liquid oscillatory culture in triangular flask. The results showed that the best carbon source for the growth of Streptomyces polychromostreptomyces (Streptomyces polychromogenes), was soluble starch, the biomass of fermentation broth was 5.8 mg/mL; yeast extract and peptone was the best nitrogen source, and the biomass was about 5.7 mg/mL.
