三峽庫(kù)區(qū)城鄉(xiāng)消落帶人工植被恢復(fù)土壤放線菌多樣性特征

【摘要】:為研究人工植被恢復(fù)后消落帶土壤放線菌群落多樣性特征,以三峽庫(kù)區(qū)典型城市消落帶重慶江北嘴大劇院消落帶和鄉(xiāng)村消落帶忠縣汝溪河流域消落帶為研究區(qū)域,于2015年6月原位采集0~20 cm、20~40 cm、40~60 cm土壤,利用限制性末端長(zhǎng)度多態(tài)性(terminal restriction fragment length polymorphism,T-RFLP)方法,對(duì)消落帶人工恢復(fù)植被土壤不同土層的放線菌群落多樣性進(jìn)行研究.結(jié)果發(fā)現(xiàn):(1)三峽庫(kù)區(qū)城鄉(xiāng)消落帶人工植被恢復(fù)后的土壤放線菌多樣性有差異,表現(xiàn)為城市消落帶土壤放線菌多樣性高于鄉(xiāng)村消落帶土壤(P0.05),特別是在表層土壤.(2)在城鄉(xiāng)消落帶中,林地土壤放線菌多樣性高于草地,且在土層之間放線菌多樣性無(wú)顯著性差異.(3)三峽庫(kù)區(qū)城鄉(xiāng)消落帶人工植被恢復(fù)后的土壤放線菌優(yōu)勢(shì)種有差異.(4)冗余分析(redundancy analysis,RDA)和蒙特卡羅置換檢驗(yàn)(Monte-Carlo permutation test)表明,城市消落帶中含水量、速效氮含量顯著影響放線菌群落多樣性;鄉(xiāng)村消落帶中土壤含水量、有機(jī)質(zhì)、速效氮和速效鉀含量顯著影響放線菌群落多樣性.結(jié)論表明在消落帶人工植被修復(fù)后,城市消落帶土壤放線菌多樣性均高于鄉(xiāng)村消落帶,這有可能是人為干擾和土壤養(yǎng)分增加所致.
[Abstract]:In order to study the diversity of soil actinomycetes community in the declination zone after the restoration of artificial vegetation, the typical urban decaying zone of the three Gorges Reservoir area, Chongqing Jiangbeizi Grand Theater, and the Zhongnian Ruxi River Basin, were used as the study areas. The diversity of actinomycetes community in different soil layers of different soil layers was studied by using restriction end length polymorphism (terminal restriction fragment length polymorphism,T-RFLP) method in 20 cm,20~40 cm,40~60 cm soil samples collected in June 2015. The results showed that: (1) the diversity of soil actinomycetes in the urban and rural areas of the three Gorges Reservoir area was higher than that in the rural areas (P0.05), and the diversity of soil actinomycetes in the urban declination zone was higher than that in the rural areas (P0.05). Especially in the surface soil. (2) the diversity of actinomycetes in woodland is higher than that in grassland in urban and rural areas. There was no significant difference in actinomycetes diversity among soil layers. (3) the dominant species of soil actinomycetes were different after the restoration of artificial vegetation in urban and rural areas of the three Gorges Reservoir area. (4) redundant analysis of (redundancy analysis,. RDA) and Monte Carlo replacement test (Monte-Carlo permutation test) showed that the water content and available nitrogen content in the urban decaying zone significantly affected the diversity of actinomycetes community. The soil moisture content, organic matter, available nitrogen and available potassium contents significantly affected the diversity of actinomycetes community. The results showed that the diversity of soil actinomycetes in the urban belt was higher than that in the rural zone after the restoration of artificial vegetation, which might be caused by human disturbance and the increase of soil nutrients.
