斑馬魚胚胎經(jīng)丙草胺暴露后對(duì)其仔魚致畸效應(yīng)的研究

【摘要】:為探討斑馬魚胚胎暴露于丙草胺后對(duì)斑馬魚仔魚的致畸效應(yīng)和致畸機(jī)理,研究了斑馬魚胚胎暴露于0.50、1.00、1.50 mg·L~(-1)丙草胺染毒液120 h后仔魚的畸形表型及比率,并通過檢測(cè)與畸形器官發(fā)育相關(guān)的基因表達(dá)探討致畸機(jī)理。結(jié)果發(fā)現(xiàn),丙草胺暴露后誘導(dǎo)斑馬魚仔魚出現(xiàn)心包囊腫、軀干彎曲、游囊關(guān)閉等畸形表型,隨著暴露濃度的增加,各畸形癥狀比例增大。1.00、1.50 mg·L~(-1)暴露組仔魚心包囊腫率分別為15.56%(P0.05)、25.56%(P0.01),脊柱彎曲率分別為27.78%(P0.01)、35.56%(P0.01);0.50、1.00、1.50 mg·L~(-1)暴露組仔魚游囊關(guān)閉率分別為20%(P0.05)、37.78%(P0.01)和60%(P0.01)。丙草胺顯著降低了心臟發(fā)育相關(guān)基因Tbx2[1.00 mg·L~(-1)(P0.01)、1.50 mg·L~(-1)(P0.05)],骨骼發(fā)育相關(guān)基因BMP-2[1.50 mg·L~(-1)(P0.05)]、BMP-4[1.00 mg·L~(-1)(P0.01)、1.50 mg·L~(-1)(P0.01)],游囊發(fā)育相關(guān)基因shha[1.50 mg·L~(-1)(P0.05)]、ihha[1.00 mg·L~(-1)(P0.05)、1.50 mg·L~(-1)(P0.05)]的表達(dá)水平。研究表明,丙草胺通過影響心臟、骨骼和游囊相關(guān)基因的表達(dá)引起斑馬魚仔魚的以上器官發(fā)育異常。
[Abstract]:In order to study the teratogenicity and teratogenic mechanism of zebrafish embryos exposed to propyloxifen, the abnormal phenotype and rate of zebrafish embryos exposed to 1.50 mg L ~ (-1) propanolamine for 120h were studied. The mechanism of teratogenicity was discussed by detecting the gene expression related to the development of malformed organs.
