花生根際產(chǎn)IAA菌的篩選鑒定及其效應研究

【摘要】:植物根際產(chǎn)IAA菌能夠分泌吲哚乙酸(IAA)促進生長,提高作物產(chǎn)量。為從砂質潮土中的花生根際篩選高產(chǎn)IAA的根際促生菌,本研究首先從長勢較好的花生根際篩選出單株菌落,對其分泌IAA能力進行定性和定量分析,并通過16S r DNA基因序列配合生理生化特征鑒定所篩選的高產(chǎn)IAA菌株,然后設置單因素試驗,探究適宜菌株生長和發(fā)酵的條件,最后通過花生盆栽試驗驗證其促生能力。結果表明,共篩選出5株產(chǎn)IAA的促生菌菌株,其中菌株HS10的IAA產(chǎn)量最高,被鑒定為特基拉芽孢桿菌。單因素試驗表明,HS10最適培養(yǎng)條件是培養(yǎng)溫度30℃,時間20 h,初始p H 8,裝液量25 m L/250 m L,碳源為果糖,氮源為硝酸鉀。盆栽試驗表明,接菌花生植株長勢明顯優(yōu)于對照,HS10對花生具有良好的促生效應。
[Abstract]:IAA-producing bacteria in the rhizosphere of plants can secrete indoleacetic acid (IAA) to promote growth and increase crop yield. In order to screen high IAA probiotic bacteria from rhizosphere of peanut in sandy tidal soil, a single colony was screened from the rhizosphere of peanut with good growth, and its IAA secretion ability was analyzed qualitatively and quantitatively. The high yield IAA strains were identified by 16s r DNA gene sequence and physiological and biochemical characteristics. Then the single factor test was set to explore the suitable conditions for the growth and fermentation of the strains. Finally, the growth promoting ability of the strains was verified by peanut pot experiment. The results showed that five strains producing IAA were screened out, among which HS10 had the highest yield of IAA and was identified as Bacillus Tequila.
