【摘要】:為了從根際土壤中篩選出具有較高溶磷能力的菌株及優(yōu)化溶磷條件,通過種子萌發(fā)初步研究溶磷促生效應(yīng)。通過稀釋涂布法分離、篩選菌株,并進(jìn)行ATB細(xì)菌鑒定儀及16S r DNA測序鑒定;采用單因子試驗(yàn)及正交試驗(yàn)優(yōu)化菌株溶磷發(fā)酵條件,利用種子發(fā)芽指標(biāo)測定菌株的促生能力。結(jié)果顯示,篩選出菌株BD-1的溶磷能力最強(qiáng),經(jīng)ATB細(xì)菌鑒定儀及16S r DNA測序,該菌株被鑒定為路德維希腸桿菌。正交試驗(yàn)結(jié)果表明,在溫度為30℃、搖床轉(zhuǎn)速為180 r/min、接種量為2 m L、初始p H值為7.0的條件下培養(yǎng)7 d的溶磷效果最好,其溶磷量為181.73 g/L。種子萌發(fā)試驗(yàn)結(jié)果表明,該溶磷菌對作物種子萌發(fā)具有一定的促進(jìn)作用。
[Abstract]:In order to screen the strains with high phosphorus solubilization ability from rhizosphere soil and optimize the phosphorus solubilizing conditions, the effect of phosphorus solubilization on growth was studied preliminarily through seed germination. The strains were isolated and screened by dilution coating method, and identified by ATB and 16s r DNA sequencing. The fermentation conditions were optimized by single factor test and orthogonal test, and the growth promoting ability of the strain was determined by seed germination index. The results showed that the strain BD-1 had the strongest phosphorus solubilizing ability. The strain was identified as Enterobacter Ludwig by ATB bacteriometer and 16s r DNA sequencing.
