【摘要】:為研究多功能菌B1514的最佳產(chǎn)孢工藝,為其擴(kuò)培、制劑的儲(chǔ)存及使用提供參考,首先測(cè)定多功能菌B1514的生長(zhǎng)曲線以初步確定其培養(yǎng)時(shí)間,然后采用單因素試驗(yàn)對(duì)影響多功能菌B1514產(chǎn)孢的接種量、初始p H值、裝液量、轉(zhuǎn)速、發(fā)酵溫度和接種種齡6個(gè)因素進(jìn)行篩選,篩選出具有顯著性影響的主效應(yīng)因素,再通過(guò)中心組合設(shè)計(jì)和響應(yīng)面分析試驗(yàn),以芽孢數(shù)量為響應(yīng)值確定各顯著影響因子的最佳水平,最后利用自動(dòng)玻璃發(fā)酵罐對(duì)優(yōu)化所得最佳培養(yǎng)條件進(jìn)行驗(yàn)證。結(jié)果表明,最佳產(chǎn)孢工藝為接種量6%,初始p H值7.6,三角瓶裝液量80 m L/500 m L,轉(zhuǎn)速235 r/min,發(fā)酵溫度36.5℃,接種種齡24 h,培養(yǎng)時(shí)間36 h,經(jīng)驗(yàn)證芽孢數(shù)量達(dá)8.19×109CFU/m L,較優(yōu)化前增加49%。
[Abstract]:In order to study the optimum sporulation technology of multifunctional bacteria B1514, and to provide reference for its expansion, storage and use, the growth curve of multifunctional bacteria B1514 was determined to determine its culture time. Then the single factor test was used to screen the inoculation amount, initial pH value, liquid loading amount, rotational speed, fermentation temperature and inoculation age of the multifunctional bacteria B1514, and the main effect factors were screened out. Through the central combination design and the response surface analysis test, the optimum level of the significant influence factors was determined with the number of spores as the response value. Finally, the optimum culture conditions were verified by using the automatic glass fermenter. The results showed that the optimum sporulation technology was inoculation volume of 6, initial pH value of 7.6, liquid volume of 80 mL / 500 mL, rotation speed of 235 r / min, fermentation temperature of 36.5 鈩,
