【摘要】：為了篩選能高效降解豬毛角蛋白的微生物,從長(zhǎng)期堆放廢棄豬毛的土壤中取樣,采用以豬毛粉為唯一碳氮源的培養(yǎng)基分離角蛋白降解菌,并對(duì)其產(chǎn)角蛋白酶條件進(jìn)行了研究。通過(guò)馴化培養(yǎng)后,篩選到5株能夠降解豬毛角蛋白的菌株,其中菌株E-2降解效果最佳。將E-2接種到以豬毛為唯一碳氮源的培養(yǎng)基中培養(yǎng)5 d后,豬毛降解率達(dá)58.6%,發(fā)酵液中角蛋白酶活為46.6 U·m L~(-1)。結(jié)合形態(tài)學(xué)觀察、生理生化特征及ITS序列分析,鑒定該菌株為解脂耶氏酵母(Yarrowia lipolytica)。E-2產(chǎn)酶影響條件實(shí)驗(yàn)結(jié)果表明,培養(yǎng)108 h后酶活性最高,產(chǎn)角蛋白酶最適條件為p H值6.0~7.0,溫度35~40℃,豬毛粉添加量15 g·L~(-1)。E-2能降解豬毛、雞毛、羊毛,但降解活性及產(chǎn)酶活性均不同。該菌株的分離篩選為微生物降解豬毛角蛋白提供了新的菌種資源,至今未見(jiàn)解脂耶氏酵母在降解毛發(fā)角蛋白方面的相關(guān)報(bào)道。
[Abstract]:In order to screen microorganisms capable of efficiently degrading porcine hair keratin, keratin degrading bacteria were isolated from soil of waste pig hair by using porcine hair powder as the sole carbon and nitrogen source, and the conditions of keratinase production were studied. After acclimation and culture, 5 strains of porcine hair keratin were screened, among which E-2 was the best. After inoculation of E-2 in the medium with pig hair as the sole carbon and nitrogen source for 5 days, the degradation rate of pig hair reached 58.6%, and the activity of keratinase in fermentation broth was 46.6 U / L ~ (-1). Combined with morphological observation, physiological and biochemical characteristics and ITS sequence analysis, the strain was identified as the optimal condition for enzyme production of Yarrowia lipolytica).E-2.
