玉米秸稈降解真菌的篩選及鑒定

【摘要】:為了篩選到高效玉米秸稈降解真菌。實(shí)驗(yàn)以樹林、稻田、玉米田等地土層下5 cm取得的腐殖質(zhì)土壤為樣品進(jìn)行富集培養(yǎng),通過剛果紅染色法進(jìn)行初步篩選,濾紙崩解法復(fù)篩,分別以羧甲基纖維素鈉和玉米秸稈粉為唯一碳源,經(jīng)30℃、120 r/min搖床培養(yǎng)72 h測(cè)定纖維素酶活性,液態(tài)發(fā)酵方法考察實(shí)際降解能力,通過分子學(xué)方法鑒定菌株。獲得2株具有較高纖維素酶活和玉米秸稈降解能力的真菌菌株Z_(2-6)和Z_(3-5),液體發(fā)酵15 d后,秸稈降解率分別是59.43%和59.13%,通過分子學(xué)鑒定都為尖孢鐮刀菌。真菌菌株Z_(2-6)和Z_(3-5)是具有較好研究?jī)r(jià)值的玉米秸稈纖維素降解真菌。
[Abstract]:In order to screen high efficiency corn straw degradation fungi. The humus soil obtained from 5 cm soil layer of forest, rice field and corn field was used as the sample for enrichment and culture. Congo red staining method was used to screen the humus soil, and filter paper disintegrating method was used to screen the humus soil.
