拮抗核盤菌Bt菌株的篩選及抑菌活性研究

【摘要】:利用平板對(duì)峙試驗(yàn)從170株Bt菌株中篩選獲得12株對(duì)核盤菌Sclerotinia sclerotiorum有抑菌活性的菌株,對(duì)其中4株活性較強(qiáng)的菌株進(jìn)行抗病相關(guān)基因分析,發(fā)現(xiàn)菌株中都含有幾丁質(zhì)酶基因,測(cè)定4AP1菌株發(fā)酵液的幾丁質(zhì)酶活力為672.4U/mL。通過對(duì)4AP1菌株pH和溫度敏感性的研究發(fā)現(xiàn),4AP1菌株最高可耐受55℃高溫及pH 12緩沖液的處理,抑菌活性與對(duì)照無顯著差異。且該菌株抑菌譜較廣,除核盤菌外,對(duì)小麥赤霉病菌Fusarium graminearum等其他9種病原真菌均有抑制作用。本研究獲得的4AP1菌株對(duì)核盤菌具有較強(qiáng)抑制作用,且耐熱耐強(qiáng)堿環(huán)境,為構(gòu)建殺蟲抗病Bt工程菌提供了菌株資源。
[Abstract]:Twelve strains with antimicrobial activity against Sclerotinia sclerotiorum were obtained from 170 BT strains by plate confrontation test. Four of them with high activity were analyzed for disease resistance related genes.
