薏苡黑穗病拮抗菌的分離鑒定及其發(fā)酵條件優(yōu)化

【摘要】:采用梯度稀釋平板涂布法從土壤中分離并純化細菌,通過平板對峙法、濾紙片法篩選出拮抗薏苡黑粉菌的細菌,經形態(tài)觀察、16S rRNA基因序列分析鑒定菌株,并優(yōu)化其發(fā)酵條件。結果表明,共分離得到拮抗菌株16個,其中拮抗菌菌株X-2對薏苡黑粉菌的抑制率相對最高,為66.7%,能夠明顯抑制病原菌的生長;經鑒定,菌株X-2為枯草芽孢桿菌(Bacillus subtilis),其最適發(fā)酵條件為溫度34℃、pH值6.0、轉速180 r/min、接種量4%、裝液量200 m L/L。
[Abstract]:The bacteria were isolated and purified from soil by gradient dilution plate coating method. The antagonistic bacteria against Coix lachrymae were screened by plate confrontation method and filter paper method. The strains were identified by morphological observation and sequence analysis of 16s rRNA gene. The results showed that 16 antagonistic strains were isolated, among which X-2 had the highest inhibition rate to Coix lachryma-black powder bacteria, which was 66.7, which could inhibit the growth of pathogenic bacteria obviously.
