高溫脅迫和抗生素對(duì)攜帶Wolbachia短管赤眼蜂生殖的影響

【摘要】:短管赤眼蜂(Trichogramma pretiosum)是膜翅目赤眼蜂科的重要天敵昆蟲,主要寄生鱗翅目害蟲的卵,在大田間有廣泛的應(yīng)用。受其體內(nèi)共生菌Wolbachia的影響,短管赤眼蜂能夠進(jìn)行完全產(chǎn)雌孤雌生殖。本論文利用高溫脅迫和抗生素對(duì)短管赤眼蜂的生殖生物學(xué)進(jìn)行了研究,獲得以下結(jié)果:1、利用Wolbachia的表面蛋白(Wolbachia Surface Protein,wsp)基因序列鑒定了所用短管赤眼蜂及其寄主米蛾(Corcyra cephalonica(Stainton))體內(nèi)的Wolbachia,它們分別屬于B超群的Kay亞群和Pip亞群。利用高溫30℃和抗生素處理短管赤眼蜂,研究這些因素對(duì)短管赤眼蜂外部形態(tài)及卵巢管的影響。結(jié)果獲得了雄性個(gè)體以及10種雌雄觸角和外生殖器不同程度組合的雌雄嵌合體。同時(shí)發(fā)現(xiàn),卵巢管變小及抱卵量減少的現(xiàn)象。論文還比較了含鹽酸四環(huán)素、利福平、慶大霉素和青霉素四種抗生素三種不同濃度蜂蜜水對(duì)短管赤眼蜂及其體內(nèi)Wolbachia的藥效作用,結(jié)果表明,對(duì)于感染W(wǎng)olbachia的短管赤眼蜂,鹽酸四環(huán)素和利福平處理比慶大霉素處理和青霉素處理更早獲得雌性后代及雌雄嵌合體,并且高濃度的抗生素(50 mg/mL、100 mg/m L)也會(huì)殺死赤眼蜂。此外,還比較了高溫脅迫(30℃)和抗生素對(duì)攜帶Wolbachia的短管赤眼蜂后代性比的影響,結(jié)果表明,抗生素處理的短管赤眼蜂雄性百分率顯著高于30℃高溫處理的后代。利福平或慶大霉素處理的F0代短管赤眼蜂的產(chǎn)卵量顯著低于青霉素處理的F0代產(chǎn)量量,到了F1、F2代,四種抗生素飼喂的短管赤眼蜂的產(chǎn)卵量并沒(méi)有差異。隨著濃度的升高,產(chǎn)卵量顯著降低。2、利用實(shí)時(shí)熒光定量PCR技術(shù)對(duì)雌雄嵌合體內(nèi)的Wolbachia進(jìn)行拷貝數(shù)檢測(cè),發(fā)現(xiàn)不同表現(xiàn)型的短管赤眼蜂體內(nèi)的Wolbachia拷貝數(shù)有顯著的差異。同時(shí),對(duì)飼喂10mg/mL鹽酸四環(huán)素蜂蜜水獲得的F1、F2、F3代雌性成蟲進(jìn)行Wolbachia進(jìn)行拷貝數(shù)檢測(cè),發(fā)現(xiàn)這三個(gè)世代雌成蟲體內(nèi)的Wolbachia拷貝數(shù)沒(méi)有顯著差異,但呈現(xiàn)逐代降低的趨勢(shì)。3、對(duì)短管赤眼蜂的轉(zhuǎn)錄組進(jìn)行測(cè)序,共得到了31439條Unigenes。其中,29439個(gè)Unigenes比對(duì)上了KOG數(shù)據(jù)庫(kù)的25個(gè)功能類別;10316條Unigenes被注釋到了183個(gè)通路上;對(duì)參與的生物過(guò)程、細(xì)胞所處的位置和基因的分子功能中得到聚類數(shù)據(jù),為進(jìn)一步分析Wolbachia對(duì)短管赤眼蜂的生殖與行為調(diào)控機(jī)制提供基礎(chǔ)。同時(shí)共得到9749個(gè)串聯(lián)重復(fù)單元,為利用微衛(wèi)星技術(shù)研究赤眼蜂的遺傳結(jié)構(gòu)或分化提供材料。
[Abstract]:Trichogramma pretiosum) is an important natural enemy insect of Hymenoptera Trichogramma, which mainly parasitizes the eggs of Lepidoptera pests and is widely used in large fields. The reproductive biology of Trichogramma brevis was studied by high temperature stress and antibiotics. The following results were obtained: 1. Wolbachia Surface protein wspsp. gene sequence of Wolbachia was used to identify Wolbachia brevis and its host, Corcyra cephalonicus Staintonia, which belong to B supergroup Kay subgroup and Pip subgroup, respectively. Treatment of Trichogramma brevis with biotin, The effects of these factors on the external morphology and ovarian duct of Trichogramma brevis were studied.
