新型擬除蟲菊酯降解酶的分子改造及降解特性

【摘要】:為了獲得穩(wěn)定性和酶活性提高的擬除蟲菊酯類降解酶,利用易錯(cuò)PCR技術(shù)對(duì)來源于海底泥宏基因組文庫的新型酯酶基因(est825)進(jìn)行體外定向進(jìn)化,獲得了一個(gè)酶活性和熱穩(wěn)定性均提高的突變酶(Est M46)。與野生型相比其酶活力提高1.5倍;最適反應(yīng)溫度提高了5℃,同時(shí)熱穩(wěn)定性明顯增強(qiáng)。此外,Est M46對(duì)氯氟氰菊酯、氯氰菊酯、氰戊菊酯和溴氰菊酯的降解率分別提升至92.21%、99.75%、93.21%和89.48%。這對(duì)降低酶的生產(chǎn)成本、促進(jìn)工業(yè)化發(fā)展和環(huán)境保護(hù)有重要意義。
