牛糞堆肥中溶解性有機物對銅綠微囊藻生理生化及產(chǎn)毒特性的影響

【摘要】:提取牛糞堆肥中的溶解性有機物(dissolved organic matter,DOM),并將DOM轉(zhuǎn)移至含有銅綠微囊藻Microcystis aeruginosa FACHB905細胞的培養(yǎng)液中,DOM含量以溶解性有機碳(DOC)質(zhì)量濃度進行衡量,分別控制培養(yǎng)液中DOC的終質(zhì)量濃度為0、15、30、60、90、120 mg/L(分別記為CK、T1、T2、T3、T4、T5)。通過測定藻細胞密度、色素含量和藻毒素含量,研究DOM對銅綠微囊藻生理生化及產(chǎn)毒特性的影響。結(jié)果表明:1)M.aeruginosa FACHB905受DOM影響,在處理后第2天,各DOM處理的細胞密度與CK相比均有所增加,T4處理的細胞密度(5.97×106個/m L)最大,比CK增加了21.67%,處理后第4天,T1、T2、T3處理均能夠促進細胞生長,T2處理的細胞密度(9.23×10~6個/m L)最大,比CK增加了15.27%,而T4、T5處理抑制細胞生長,處理后第6天,僅T1及T2處理能夠促進細胞生長,T1處理的細胞密度(13.34×10~6個/m L)最大,比CK增加了10.35%,而T3、T4、T5處理抑制細胞生長;2)堆肥來源DOM能夠抑制藻毒素的合成,單位細胞藻毒素Microcystin LR含量在處理后第2天T5處理的最低,比CK下降了53.13%,而Dha7 Microcystin LR含量在處理后第4天T5處理的最低,與CK比較下降了78.51%;3)各DOM處理對單位細胞內(nèi)Chl a和Carotenoid的合成沒明顯影響。
[Abstract]:Dissolved organic matter (dissolved organic matterus) was extracted from cow manure compost, and DOM was transferred to the culture medium containing Microcystis aeruginosa FACHB905 cells. The content of Dom was measured by the mass concentration of dissolved organic carbon (DOC). The final mass concentration of DOC in culture medium was controlled to be 0 1530g / L, 6090g / L, respectively. The effects of DOM on physiological, biochemical and toxic properties of microcystis aeruginosa were studied by determining the cell density, pigment content and phycotoxin content of microcystis aeruginosa, respectively. The results showed that DOM was affected by DOM, and the results showed that the microcystis aeruginosa FACHB905 was affected by DOM, and the cell density, pigment content and phycotoxin content of microcystis aerginosa were determined. On the second day after treatment, the cell density of each DOM treatment was 5.97 脳 106 / mL, which was higher than that of CK, and the cell density of T4 treatment was 5.97 脳 106 / mL. On the 4th day after treatment, the cell density of T 2 treatment increased by 9.23 脳 10 ~ 6 / mL, and increased 15.27% than CK, while T4 + T _ 5 inhibited cell growth, and on the 6th day after treatment, the cell density was increased by 9.23 脳 10 ~ 6 / mL. Only T 1 and T 2 treatments could promote cell growth. The cell density of T 1 treatment was 13.34 脳 10 ~ 6 / mL, which was 10.35% higher than CK, while T _ 3 T _ 4 T _ 4 T _ 5 treatment inhibited cell growth. DOM, a compost source, could inhibit the synthesis of phycotoxin. The content of Microcystin LR was the lowest in T5 treatment on the 2nd day after treatment, which was 53.13% lower than that of CK, while the lowest in T5 treated with Dha7 Microcystin on the 4th day after treatment. Compared with CK, there was no significant effect of DOM treatment on the synthesis of Chl a and Carotenoid in the unit cells.
