低溫適應(yīng)型植物根際促生細(xì)菌的篩選及促生效應(yīng)研究

【摘要】:[目的]優(yōu)良菌種是微生物肥料的基礎(chǔ),低溫適應(yīng)型菌株對(duì)于研制在低溫條件下應(yīng)用的微生物肥料具有重要意義,本研究基于菌株的低溫適應(yīng)性篩選植物根際促生菌。[方法]利用稀釋涂布的分菌方法從土壤樣品中分離出快速生長(zhǎng)的根際促生菌,并篩選在低溫條件下生長(zhǎng)良好的菌株,進(jìn)一步分析所篩菌株的促生特性,通過盆栽試驗(yàn)驗(yàn)證促生效果,并研究微生物肥料對(duì)土壤酶活性的影響,探究所篩菌株的促生機(jī)制。[結(jié)果]初篩所得36株菌株均具有溶磷、產(chǎn)NH3、產(chǎn)吲哚乙酸(IAA)及產(chǎn)嗜鐵素等一種或多種促生性能,其中具有產(chǎn)氨能力的14株,占所篩菌株的38.9%;產(chǎn)IAA的20株,占所篩菌株的55.6%;產(chǎn)鐵載體的12株,占所篩菌株的33.3%;具有溶解有機(jī)磷能力的15株,占所篩菌株的41.7%。通過低溫生長(zhǎng)驗(yàn)證,8株菌可以在低溫(15℃)下生長(zhǎng),均具有產(chǎn)NH3和產(chǎn)IAA的促生特性,菌株D4的IAA產(chǎn)量最大,達(dá)到162 mg·L~(-1)。結(jié)合盆栽促生試驗(yàn)結(jié)果,最終選擇D4和R1兩株芽孢桿菌制作生物有機(jī)肥在冬季大棚的低溫條件下進(jìn)行肥效試驗(yàn)。結(jié)果表明生物有機(jī)肥對(duì)青菜的株高、根長(zhǎng)和鮮質(zhì)量的影響均高于普通有機(jī)肥和化肥。土壤酶活性測(cè)定結(jié)果表明,生物有機(jī)肥與化肥混施處理使土壤脲酶、蔗糖酶、過氧化氫酶和堿性磷酸酶活性顯著增加;16S rRNA基因進(jìn)行序列同源性分析,菌株D4為巨大芽孢桿菌,菌株R1為短小芽孢桿菌。[結(jié)論]所篩選得到的低溫適應(yīng)型根際促生菌和研制的生物肥料,具有在寒冷地區(qū)農(nóng)業(yè)生產(chǎn)中應(yīng)用的潛力。
[Abstract]:[objective] excellent bacteria species are the basis of microbial fertilizer, and low temperature adapted strains are of great significance for the development of microbial fertilizer under low temperature conditions. In this study, rhizosphere growth promoting bacteria were screened based on the low temperature adaptability of strains. [methods] Rapid growing rhizosphere promoting bacteria were isolated from soil samples by dilution coating method, and strains growing well at low temperature were screened. The effects of microbial fertilizer on soil enzyme activity were studied, and the growth promoting mechanism of the screening strain was studied. [results] the 36 strains obtained from the primary screening all had phosphorus solubilization, and the effect of microbial fertilizer on soil enzyme activity was studied. Among them, 14 strains with ammonia production capacity, accounting for 38.9% of the screened strains, 20 strains producing IAA, accounting for 55.6% of the screened strains, 12 strains of iron carriers, 15 strains with ability to dissolve organophosphorus, 41.7% of the screened strains. It was proved by low temperature growth that 8 strains could grow at 15 鈩,
