數(shù)值研究血管內(nèi)白細胞介導(dǎo)的黑色素瘤細胞粘附

【摘要】:該文研究了中性粒白細胞介導(dǎo)腫瘤細胞粘附于血管內(nèi)皮細胞的過程。以血液中的黑色素瘤細胞為例,考慮其與白細胞之間特殊的分子配對成鍵以及表面摩擦因素,采用準(zhǔn)定常方法,數(shù)值模擬了黑色素瘤細胞在白細胞表面運動粘附的過程。在不同的來流剪切率下,黑色素瘤細胞在與白細胞接觸過程中的動力學(xué)特性會有不同。由于這種生化鍵的存在,增加了黑色素瘤細胞與白細胞的接觸時間。數(shù)值結(jié)果表明:在低來流剪切率時,黑色素瘤細胞與白細胞的接觸時間增加了1倍;在高來流剪切率時,黑色素瘤細胞雖然會在某一位置脫離白細胞表面,但是他們的接觸時間還是延長了58.6%。說明這種生化鍵的存在極大促進黑色素瘤細胞的粘附,提高了腫瘤轉(zhuǎn)移的幾率。
[Abstract]:The process of neutrophil mediated tumor cell adhesion to vascular endothelial cells was studied. Taking melanoma cells in blood as an example, the special molecular pairing bonding and surface friction factors between melanoma cells and leukocytes were considered. A quasi-stationary method was used to numerically simulate the adhesion of melanoma cells on the surface of white blood cells. The kinetic characteristics of melanoma cells in the process of contact with white blood cells will be different. Due to the existence of this biochemical bond, the contact time between melanoma cells and white blood cells is increased. The numerical results show that at low flow shear rate, The contact time between melanoma cells and white blood cells doubled; at high flow shear rates, melanoma cells may escape from the surface of white blood cells at a certain location. However, their exposure time was prolonged by 58.6%, indicating that the existence of this biochemical bond greatly promoted the adhesion of melanoma cells and increased the risk of tumor metastasis.
