分離自安徽銅陵酸性礦山環(huán)境的AMDY2-9-2和AMDSY3-6-1還原鐵的研究

【摘要】:本論文對(duì)分離自安徽銅陵酸性礦山環(huán)境中的兩株鐵還原菌進(jìn)行了生物學(xué)鑒定和還原鐵研究。通過對(duì)安徽銅陵酸性礦山環(huán)境中的野外樣品(酸礦水、尾砂、排水溝底泥)進(jìn)行采集,用選擇性培養(yǎng)基FeT培養(yǎng)樣品,經(jīng)過分離純化獲得兩株鐵還原微生物AMDY2-9-2和AMDSY3-6-1。對(duì)兩株微生物的16SrDNA測序和同源性分析表明,菌株AMDY2-9-2與A cidithiobacillus thiooxidans親緣關(guān)系最為接近,同源性為99.79%,而AMDSY3-6-1與Acidithiobacillus ferridurans親緣關(guān)系最為接近,同源性為95.78%。菌株AMDY2-9-2和AMDSY3-6-1在FeT培養(yǎng)基上生長特征為:兩株細(xì)菌均能形成0.05-0.1 mm的圓形菌落,菌落呈白色,透明狀,其邊緣齊整。兩株細(xì)菌均為革蘭氏陰性的好氧菌,菌體大小(長×寬):0.5-1.0μm×0.2-0.4 μm。兩株細(xì)菌均能夠利用苯丙氨酸和硫化氫,對(duì)尿素的利用呈弱陽性,但是對(duì)葡萄糖、乳糖、麥芽糖、甘露醇、甘露糖、蔗糖、阿拉伯糖、阿拉伯醇、D-核糖、木糖、木糖醇、半乳糖、松二糖、松三糖、山梨醇、山梨糖、蜜二糖、纖維二糖、葡萄糖銨、甲基-D葡萄糖苷、果糖、血清菊糖、DNA、酒石酸鹽、葡萄糖酸鹽、醋酸鹽、衛(wèi)茅醇(甜醇)、苦杏仁苷、鼠李糖、糊精、淀粉、肌醇、水楊素、β-半乳糖苷、丙二鹽酸、西蒙氏枸櫞酸鹽、七葉苷、乙酰胺、硝酸鹽(還原)、馬尿酸鹽、N-乙酰葡萄糖胺、丙酮酸鈉等化合物不能夠利用,不具有鳥氨酸脫羧酶、賴氨酸脫羧酶、精氨酸脫羧酶活性。在pH 2.0-11.0的范圍兩株細(xì)菌都能夠生長,但是AMDY2-9-2最適生長pH值為5.0,AMDSY3-6-1最適生長pH值為6.0;兩株細(xì)菌最適生長溫度均為30℃,而且在NaCl含量為0%的FeT培養(yǎng)基中生長最好。AMDY2-9-2和AMDSY3-6-1與鐵離子的作用機(jī)制研究表明,對(duì)于可溶性三價(jià)鐵離子,兩株細(xì)菌均需要完整的活細(xì)胞才能實(shí)現(xiàn)鐵離子還原過程,單獨(dú)作用的胞內(nèi)活性酶、滅活的細(xì)胞和滅活的胞內(nèi)酶均不能完成鐵離子還原過程。兩株細(xì)菌在不同三價(jià)鐵離子濃度的FeT培養(yǎng)液中培養(yǎng)發(fā)現(xiàn),與菌株AMDSY3-6-1比,菌株AMDY2-9-2生長比較快;對(duì)菌體與鐵帽的相互作用研究結(jié)果表明,兩株細(xì)菌均表現(xiàn)出還原速度慢的特點(diǎn),詳細(xì)的鐵還原機(jī)制值得進(jìn)一步研究。
[Abstract]:In this paper, two iron reducing bacteria isolated from the environment of Tongling acid mine in Anhui province were identified and studied. Two iron reducing microorganism (AMDY2-9-2 and AMDSY3-6-1) were isolated and purified from acidic mine environment in Tongling, Anhui Province by collecting the samples (acid ore water, tailings and draining ditch sediment), and using FeT culture medium to isolate and purify the two strains of iron reducing microorganism AMDY2-9-2 and AMDSY3-6-1. The 16SrDNA sequencing and homology analysis of the two microbes showed that the AMDY2-9-2 and A cidithiobacillus thiooxidans were the most close, the homology was 99.79, while AMDSY3-6-1 and Acidithiobacillus ferridurans were the most close, and the homology was 95.78. The growth characteristics of strain AMDY2-9-2 and AMDSY3-6-1 on FeT medium were as follows: both of them could form round colony of 0.05-0.1 mm, the colony was white and transparent, and its edge was even. The two strains were Gram-negative aerobic bacteria, and the cell size (length 脳 width) was 0.5-1.0 渭 m 脳 0.2-0.4 渭 m. Both strains were able to use phenylalanine and hydrogen sulfide and were weakly positive for urea use, but for glucose, lactose, maltose, mannitol, mannose, sucrose, arabinol D-ribose, xylitol, Galactose, pine disaccharide, trisaccharide, sorbitol, honey disaccharide, fibronectose, ammonium glucose, methyl D glucoside, fructose, serum chrysanthemum DNA, tartrate, gluconate, acetate, citronol (steviol), amygdalin, Rhamnose, dextrin, starch, inositol, salicylate, 尾 -galactoside, propanedioic acid, Simon's citrate, aescine, acetamide, nitrate (reduction), hippate N-acetyl glucosamine, sodium pyruvate, etc. No ornithine decarboxylase, lysine decarboxylase, arginine decarboxylase activity.
