應(yīng)用抑制性差減雜交技術(shù)研究棗縮果病病程相關(guān)基因

【摘要】:【目的】了解細(xì)交鏈孢菌侵染棗果實(shí)過程的差異表達(dá)基因!痉椒ā恳园资炱凇涿酃蕖瘲椀墓麑(shí)為材料,人工接種細(xì)交鏈孢菌,分別在接種0.5、1、2、3和4 d后取樣,通過抑制性消減雜交技術(shù)(suppression subtractive hybridization,SSH)構(gòu)建了病原菌誘導(dǎo)的差異表達(dá)的c DNA消減文庫。【結(jié)果】PCR鑒定隨機(jī)挑取的陽性克隆,顯示插入片段大小為200~900 bp。隨機(jī)挑選200個(gè)陽性克隆進(jìn)行測序,利用BLASTx在Gen Bank Nr數(shù)據(jù)庫進(jìn)行序列比對分析,共獲得118個(gè)Unigenes。對這些ESTs進(jìn)行功能分類發(fā)現(xiàn)細(xì)交鏈孢菌侵染下誘導(dǎo)表達(dá)基因涉及植物細(xì)胞內(nèi)的多種代謝和應(yīng)答過程,其中包括抗病/防御類、信號(hào)傳導(dǎo)途徑類、新陳代謝類、蛋白質(zhì)合成和加工類及細(xì)胞結(jié)構(gòu)的組成等,尤其以抗病/防御類基因所占比例最大(27.17%)。【結(jié)論】通過SSH研究了縮果病病原侵染果實(shí)過程的差異表達(dá)基因,鑒定到一些與棗縮果病相關(guān)的基因,為進(jìn)一步研究相關(guān)基因及今后篩選防治棗縮果病提供理論基礎(chǔ)。
