氯霉素對(duì)費(fèi)氏弧菌0~24h的Hormesis效應(yīng)及其機(jī)制初探

【摘要】:為了探究抗生素對(duì)發(fā)光菌的Hormesis效應(yīng),選擇費(fèi)氏弧菌(Vibrio fischeri,V.fischeri)為受試生物,氯霉素為研究對(duì)象,測(cè)定了0~24h下氯霉素對(duì)V.fischeri的發(fā)光強(qiáng)度(Hv)及生長(zhǎng)量(OD_(600))的作用,探討氯霉素對(duì)V.fischeri的低濃度促進(jìn)效應(yīng)及其可能機(jī)制.結(jié)果表明,氯霉素對(duì)V.fischeri的發(fā)光強(qiáng)度具有明顯的促進(jìn)作用,且僅出現(xiàn)在細(xì)菌生長(zhǎng)的延滯期.根據(jù)V.fischeri的發(fā)光機(jī)制,結(jié)合Gaussian量子化學(xué)計(jì)算結(jié)果,即氯霉素的最正氫電荷為0.244,遠(yuǎn)大于煙酰胺腺嘌呤二核苷酸(NADH)的最正氫電荷0.135,提出氯霉素通過提供質(zhì)子促進(jìn)熒光反應(yīng),進(jìn)而產(chǎn)生Hormesis效應(yīng)的可能機(jī)制.
[Abstract]:In order to investigate the Hormesis effect of antibiotics on luminescent bacteria, Vibrio flexneri (Vibrio fischeri,V.fischeri) was selected as the tested organism and chloramphenicol was used as the research object. The effects of chloramphenicol on V.fischeri luminescence intensity (Hv) and growth capacity (OD_ (600) at 0 ~ 24 h were measured. The effects of chloramphenicol on the low concentration of V.fischeri and its possible mechanism were discussed. The results showed that chloramphenicol promoted the luminescence intensity of V.fischeri obviously, and only appeared in the lag period of bacterial growth. According to the luminescence mechanism of V.fischeri and the results of Gaussian quantum chemistry calculation, the most positive hydrogen charge of chloramphenicol is 0.244, which is much larger than that of nicotinamide adenine dinucleotide (NADH) 0.135. The possible mechanism of chloramphenicol (chloramphenicol) promoting fluorescence reaction by providing proton to produce Hormesis effect is put forward.
