抗鋅細(xì)菌ZS2的分離鑒定及抗金屬特性研究

【摘要】:篩選可有效降解礦區(qū)土壤重金屬的抗性細(xì)菌,在土壤環(huán)境的生物修復(fù)中應(yīng)用前景廣闊。從陜西商洛鉛鋅礦重金屬污染土壤中篩選獲得一株對重金屬鋅有高抗性的菌株,命名為ZS2。通過形態(tài)特征、生理生化特性及16S r RNA序列分析,初步鑒定該菌為不解糖假蒼白桿菌(Pseudochrobactrum asaccharolyticum)。研究其抗重金屬及重金屬吸附特性,結(jié)果表明:ZS2對Zn~(2+)具有較高抗性,最大耐受濃度20 mmol/L,且對多種重金屬(Pb~(2+)、Cu~(2+)、Cr6+和Cd~(2+))也具有耐受性;對低濃度Zn~(2+)去除效果最佳,Zn~(2+)濃度為2.0 mmol/L時吸附率高達(dá)55.25%;同時對高濃度Zn~(2+)仍具有去除作用。不解糖假蒼白桿菌ZS2是鮮有的抗重金屬假蒼白桿菌屬細(xì)菌,具有優(yōu)良的修復(fù)土壤重金屬污染應(yīng)用價值。
[Abstract]:Screening resistant bacteria that can effectively degrade heavy metals in mining areas has a bright future in bioremediation of soil environment. A strain with high resistance to heavy metal zinc was obtained from heavy metal contaminated soil of Shangluo lead-zinc mine in Shaanxi Province. It was named ZS2.. By morphological, physiological and biochemical characteristics and 16s r RNA sequence analysis, the strain was identified as (Pseudochrobactrum asaccharolyticum). The results showed that ZS2 had high resistance to Zn~ (2), maximum tolerance concentration of 20 mmol/L, and tolerance to many heavy metals (Pb~ (2), Cu~ (2), Cr6 and Cd~ (2). The removal efficiency of low concentration Zn~ (2) was the best, the adsorption rate of Zn~ (2) was up to 55.25 mmol/L, and the removal of high concentration of Zn~ (2) was still effective. Pseudopallidobacillus saccharum (ZS2) is a rare bacteria resistant to heavy metal, which has good application value in remediation of heavy metal pollution in soil.
