以預(yù)處理污泥為底物生產(chǎn)微生物絮凝劑的研究

【摘要】:生物絮凝劑因其自身優(yōu)點(diǎn)及瓶頸問題,使得它在污水凈化生產(chǎn)實(shí)踐一直受到廣泛關(guān)注,而培養(yǎng)產(chǎn)絮菌的基質(zhì)價(jià)格貴問題是妨礙絮凝劑大規(guī)模應(yīng)用于凈化污水和工業(yè)生產(chǎn)的最重要原因。本課題就絮凝劑產(chǎn)生菌發(fā)酵培養(yǎng)成本較高問題提出新的思路,以污泥處理后的提取液作為原料制備污泥提取液發(fā)酵培養(yǎng)基,對(duì)發(fā)酵過程中的環(huán)境因子進(jìn)行考察,最后對(duì)這一研究進(jìn)行了綜合效益評(píng)估。對(duì)污泥進(jìn)行不同條件(不同溫度、時(shí)間、酸堿度)的預(yù)處理,分析其提取液的TOC濃度、TN濃度、TP濃度等,從而確定最佳的預(yù)處理?xiàng)l件。絮凝劑產(chǎn)生菌LLin6直接利用污泥提取液發(fā)酵產(chǎn)絮效果并不理想,經(jīng)過調(diào)節(jié)濃度和補(bǔ)加物質(zhì),確定20~25 g/L的污泥在p H=12條件下處理3 h,后經(jīng)離心去除沉淀留取上清液備用,調(diào)節(jié)p H值至7.0左右,添加8 g/L的葡萄糖,112℃滅菌30 min后接入絮凝劑產(chǎn)生菌LLin6進(jìn)行發(fā)酵培養(yǎng),24 h發(fā)酵后可以獲得85%以上的絮凝率。對(duì)絮凝劑產(chǎn)生菌LLin6在所配制的污泥提取液培養(yǎng)基中生長狀況進(jìn)行分析發(fā)現(xiàn),產(chǎn)絮菌LLin6在污泥提取液培養(yǎng)基中生長狀況良好。以污泥提取液作為培養(yǎng)基原料,對(duì)絮凝劑產(chǎn)生菌LLin6在其中產(chǎn)絮發(fā)酵進(jìn)行考察。在四組單因素實(shí)驗(yàn)基礎(chǔ)上,利用響應(yīng)曲面法預(yù)測絮凝劑產(chǎn)生菌LLin6在污泥提取液中最佳發(fā)酵條件:發(fā)酵溫度24.7℃、培養(yǎng)基初始p H為6.5、接種量為6%、發(fā)酵時(shí)間22 h,此條件下理論上可以獲得的絮凝率為87.14%。而實(shí)際絮凝率為85.54%,與預(yù)測值相接近,證明響應(yīng)曲面分析法得到的發(fā)酵條件可信度高,可以預(yù)測響應(yīng)值。提出產(chǎn)絮菌LLin6以污泥提取液為底物生產(chǎn)微生物絮凝劑的工藝流程,對(duì)以處理后的剩余污泥為底物制備生物絮凝劑的產(chǎn)品進(jìn)行了成本分析,產(chǎn)3.5T液體生物絮凝劑,可資源化利用剩余污泥(濃度一般為6 g/L)約14.6 T,生物絮凝劑原料成本由730元降低了404.5元,相當(dāng)節(jié)約了的55%以上,大幅降低了原料成本。除了經(jīng)濟(jì)效益,還有它隨之帶來的社會(huì)和環(huán)境效益。
[Abstract]:Because of its advantages and bottlenecks, bioflocculants have been widely concerned in the production of sewage purification. The most important reason for the large-scale application of flocculant in wastewater purification and industrial production is the expensive substrate price of flocculant. In this paper, a new idea about the high cost of flocculant-producing bacteria fermentation was put forward. The sludge extract was used as the raw material to prepare the fermentation medium of sludge extract, and the environmental factors in the process of fermentation were investigated. Finally, the comprehensive benefit evaluation of this study is carried out. The sludge was pretreated with different conditions (different temperature, time, pH), and the TOC concentration, TN concentration and TP concentration of the extract were analyzed, so as to determine the best pretreatment conditions. The flocculant producing strain LLin6 directly used sludge extract to ferment flocculating effect is not ideal. After adjusting the concentration and adding substance, the sludge of 20 ~ 25 g / L was treated at pH 12 for 3 h, and then centrifugally removed and precipitated to take the supernatant.
