酪氨酸酶固定化碳材料對苯酚的生物降解性能

【摘要】:以新型碳材料氧化石墨烯作為載體,利用共價結(jié)合方法制備酪氨酸酶固定化氧化石墨烯復(fù)合物,并利用該復(fù)合物對苯酚進(jìn)行催化降解,探討酶的加載特性和酶的催化活性,以及固定化酪氨酸酶催化降解苯酚的最優(yōu)條件及儲存穩(wěn)定性。通過對固定化酪氨酸酶進(jìn)行活性和固定量分析后認(rèn)為:單位質(zhì)量載體的酶固定量為1.78 mg/mg,單位質(zhì)量載體的酶活性為1 880.6 U/mg;固定化酪氨酸酶在30h內(nèi)對47.06mg/L苯酚的降解率可達(dá)86.3%,降解反應(yīng)的最優(yōu)條件為pH=7.0、溫度=25℃;固定化酪氨酸酶在4℃條件下30d后仍保持初始活性的77.7%,其穩(wěn)定性優(yōu)于游離酪氨酸酶。另外,在氧化石墨烯上引入磁顆粒,既簡化了酶固定流程,又能做到回收利用。
[Abstract]:The graphene oxide complex immobilized by tyrosinase was prepared by covalent binding method with a new carbon material graphene oxide as the carrier. The complex was used to catalyze the degradation of phenol. The loading characteristics and catalytic activity of the enzyme were discussed. The optimal conditions and storage stability of phenol degradation catalyzed by immobilized tyrosinase were also discussed. After quantitative analysis of immobilized tyrosinase activity and fixation analysis, it is concluded that the immobilized amount of the immobilized tyrosinase is 1.78 mg / mgand the enzyme activity of the unit mass carrier is 1 880.6 Umg.The degradation rate of phenol by immobilized tyrosinase within 30 hours is 47.06 mg / L phenol.
