耐受重金屬鉛乳酸菌的分離篩選及鑒定

【摘要】:通過對包頭尾礦庫區(qū)、白云鄂博采礦區(qū)重金屬污染嚴重區(qū)域土壤樣品中的乳酸菌進行分離、篩選,共得到62株乳酸菌,并對分離得到的乳酸菌進行耐受鉛離子的測定,62株乳酸菌對鉛離子均有一定的耐受能力,不同菌株對鉛離子的耐受能力有所不同;鉛離子濃度為1200~9600 mg/L時,隨著培養(yǎng)時間的增長,菌株對耐受鉛離子的修復能力逐漸增強;30℃下分離得到的菌株對鉛離子的修復能力明顯優(yōu)于37℃下分離得到的菌株;菌株BY-Fa4、BG-5b3、BG-8b2等13株菌可在鉛離子濃度為7200 mg/L的環(huán)境下生長,菌株BY-Fa1、BY-Fc2、BG-6b1可在鉛離子濃度為8400 mg/L的環(huán)境下生長。實驗結果得出篩選得到的13株乳酸菌對鉛離子不僅具有較強的耐受能力,而且還具有較強的修復能力。對篩選出13株高耐受鉛離子的菌株進行16S rRNA序列分析,鑒定結果為戊糖片球菌2株、清酒乳桿菌1株、食竇魏斯氏菌5株、戊糖乳桿菌2株、植物乳桿菌3株。
[Abstract]:A total of 62 strains of lactic acid bacteria were isolated from soil samples in Baotou tailing area and heavy metal polluted area of Baiyunebo mining area, and 62 strains of lactic acid bacteria were obtained. The results showed that 62 strains of lactic acid bacteria had certain tolerance to lead ion, and different strains had different tolerance ability to lead ion, and when lead ion concentration was 1 200 ~ 9 600 mg/L, the tolerance ability of different strains to lead ion increased with the increase of culture time.
