【摘要】：塔瑪亞歷山大藻是一種有毒甲藻,常引發(fā)赤潮,嚴(yán)重威脅海洋生態(tài)的穩(wěn)定和人類的健康,細(xì)菌Deinococcus sp.Y35表現(xiàn)出對塔瑪亞歷山大藻的殺藻能力,為促進(jìn)菌株生長、提高殺藻效果并方便保存,對菌株Y35培養(yǎng)條件進(jìn)行優(yōu)化,并制備殺藻菌劑.分別確定菌株Y35生長所需的最適氮源、碳源、無機(jī)鹽,并確定其最適添加量,在優(yōu)化的基礎(chǔ)上完成凍干菌劑的制備和最適凍干保護(hù)劑的選擇.菌株Y35生長的最適培養(yǎng)基成分是1.0%胰蛋白胨和0.5%酵母粉.在優(yōu)化的培養(yǎng)基基礎(chǔ)上對菌株Y35進(jìn)行發(fā)酵,培養(yǎng)至對數(shù)期后進(jìn)行冷凍干燥,制備殺藻菌劑.菌株Y35需要添加1.0 g/L的蔗糖作為凍干保護(hù)劑.殺藻菌劑的殺藻添加量為2.0 mg/m L.本研究可為下一步將細(xì)菌應(yīng)用于赤潮治理奠定基礎(chǔ).
[Abstract]:Alexandrium tamarense is a kind of toxic dinoflagellate, which often causes red tide, which seriously threatens the stability of marine ecology and human health. Bacterial Deinococcus sp.Y35 shows the ability of killing algal to Alexandrium tamarense, in order to promote the growth of the strain. The culture conditions of strain Y35 were optimized and algaecide was prepared. The optimum nitrogen source, carbon source and inorganic salt for the growth of strain Y35 were determined, and the optimum addition amount was determined. On the basis of optimization, the preparation of lyophilized bacteria agent and the selection of optimal lyophilized protectant were completed. The optimum medium for growth of strain Y35 was 1.0% tryptone and 0.5% yeast powder. Strain Y35 was fermented on the basis of optimized culture medium and then freeze-dried in logarithmic period to prepare algaecide. Strain Y35 needs 1.0 g / L sucrose as freeze-drying protectant. The addition of algaecide was 2.0 mg/m / L. This study can lay a foundation for the application of bacteria in red tide control.
