【摘要】:CD-like微流控芯片是一種簡單高效的生化分析平臺,它通過調(diào)整自身的轉(zhuǎn)速實現(xiàn)對芯片內(nèi)部微流體的操作和控制,具有試劑用量少、集成度高、分析速度快、體積小等優(yōu)點。本論文為實現(xiàn)CD-Like微流控芯片多指標、多波長的探測目的,圍繞芯片的光探測系統(tǒng)設計,展開了以下研究工作:1.采用閃爍氙燈作為光源,微型光譜儀作為檢測器,設計并搭建了光探測系統(tǒng)。驗證了多路光電檢測電路,給出了芯片精確定位與多參數(shù)檢測方法;2.對影響光探測系統(tǒng)性能的主要因素進行了分析研究,給出了誤差源,建立了光源輸出穩(wěn)定性、雜散光與吸光度誤差之間的關系;給出了以雙波長檢測為主體的光源輸出抖動補償方法并對其進行了實驗驗證,相比單波長檢測方法,檢測結(jié)果的重復性提高了18倍;3.以葡萄糖、總蛋白、白蛋白和二氧化碳結(jié)合力等生化指標的檢測為目標,對光探測系統(tǒng)性能進行了研究。測試結(jié)果顯示,定標曲線的線性度達到99%以上,吸光度線性范圍、重復性和雜散光均滿足國家標準。本論文的工作將有利于推進CD-Like微流控芯片在醫(yī)學臨床領域的應用,對實現(xiàn)生化分析儀器的集成化與小型化具有重要意義。
[Abstract]:CD-like microfluidic chip is a simple and efficient biochemical analysis platform. It can operate and control the microfluid inside the chip by adjusting its rotational speed. It has the advantages of less reagent consumption, high integration, fast analysis speed and small volume. In order to realize the multi-target and multi-wavelength detection of CD-Like microfluidic chip, the following research work has been carried out in this paper, focusing on the design of the optical detection system of the chip. The light detection system is designed and built by using flash xenon lamp as light source and micro spectrometer as detector. The multichannel photoelectric detection circuit is verified, and the accurate location of the chip and the method of multi-parameter detection are given. The main factors affecting the performance of the optical detection system are analyzed and studied. The error source is given, and the relationship between the output stability of the light source, the stray light and the absorbance error is established. A method for compensating the output jitter of the light source with dual wavelength detection as the main part is presented and verified experimentally. The repeatability of the detection results is increased by 18 times and 3 times than that of the single wavelength detection method. The performance of the optical detection system was studied based on the detection of biochemical indexes such as glucose, total protein, albumin and carbon dioxide. The results show that the linearity of calibration curve is over 99%, and the linear range of absorbance, repeatability and stray light are all up to the national standard. The work of this paper will be helpful to promote the application of CD-Like microfluidic chip in the field of medical clinic, and it is of great significance to realize the integration and miniaturization of biochemical analysis instruments.
