表面復(fù)合修飾納米超順磁性材料固定化α-淀粉酶性能研究

【摘要】:以復(fù)合修飾的納米超順磁性Fe3O4顆粒聚集體為載體固定化α-淀粉酶,比較分析固定化α-淀粉酶及游離α-淀粉酶的酶學(xué)性能。研究固定化及游離α-淀粉酶的最適溫度、最適p H、操作穩(wěn)定性及基本動(dòng)力學(xué)等。結(jié)果表明,固定化α-淀粉酶最適p H為7,最適溫度為60℃。固定化α-淀粉酶與游離α-淀粉酶相比,具有更好的溫度和酸堿的耐受性。固定化α-淀粉酶重復(fù)催化反應(yīng)10次,相對酶活力仍剩余72.09%,重復(fù)操作的半衰期為18.97次,具有良好的操作穩(wěn)定性。固定化α-淀粉酶的米氏常數(shù)Km值為45.31 mg/m L,親和性弱于游離α-淀粉酶。
[Abstract]:The immobilized 偽 -amylase was immobilized on the composite modified nano-superparamagnetic Fe3O4 particle aggregates, and the enzymatic properties of the immobilized 偽 -amylase and free 偽 -amylase were compared and analyzed.
