持續(xù)與間歇跑臺(tái)運(yùn)動(dòng)對(duì)大鼠血清及不同組織自由基代謝及抗氧化能力的比較研究

【摘要】:研究目的:探討6周持續(xù)與間歇跑臺(tái)運(yùn)動(dòng)對(duì)大鼠血清及不同組織(心肌,肝臟,股四頭肌,腓腸肌,比目魚肌)MDA含量變化及SOD、T-AOC活性變化的影響。研究方法:雄性SD大鼠24只,隨機(jī)分為安靜對(duì)照組(S組)、持續(xù)訓(xùn)練組(C組)和長時(shí)間歇訓(xùn)練組(I組),共3組,每組8只。運(yùn)動(dòng)組先進(jìn)行為期1周的適應(yīng)性跑臺(tái)。適應(yīng)性跑臺(tái)訓(xùn)練按照以下3級(jí)負(fù)荷進(jìn)行:(1)第1-2天以15m/min×20min;(2)第3-4天18m/min×20min;(3)第5-6天21m/min×20min。正式訓(xùn)練時(shí),安靜組不運(yùn)動(dòng),正常飼養(yǎng),持續(xù)6周。運(yùn)動(dòng)組第一周以22m/min的速度開始進(jìn)行跑臺(tái)訓(xùn)練,以后每周以1m/min的速度遞增,直至第6周27m/min為止,6天/周。其中持續(xù)訓(xùn)練組每天訓(xùn)練一次,訓(xùn)練40min;長時(shí)間歇組每天訓(xùn)練2組,每組訓(xùn)練20min,組間休息3h。6周訓(xùn)練結(jié)束后不限飲水,禁食24h,處死分別取出心尖血5ml,心肌,肝臟,股四頭肌,比目魚肌,腓腸肌等組織置于-80℃冰箱待測(cè),全程在冰上進(jìn)行,最后采用比色法(試劑盒)測(cè)定大鼠的MDA、SOD、T-AOC等指標(biāo)。研究結(jié)果:1.MDA變化:6周持續(xù)與間歇跑臺(tái)運(yùn)動(dòng)后,持續(xù)運(yùn)動(dòng)組血清MDA含量為5.7±0.49nmol/ml,與安靜對(duì)照組相比顯著降低(P0.05),長時(shí)間歇運(yùn)動(dòng)組血清MDA含量為3.65±0.68nmol/ml,與安靜組相比有著極顯著的差異(P0.01),且長時(shí)間歇組與持續(xù)訓(xùn)練組相比呈現(xiàn)出了顯著差異(P0.05);心肌MDA含量持續(xù)訓(xùn)練組與安靜對(duì)照組相比無明顯差異(P0.05),長時(shí)間歇組與安靜對(duì)照組相比,差異顯著(P0.05),長時(shí)間歇組與持續(xù)組相比,顯著下降(P0.05);肝臟MDA含量長時(shí)間歇組與安靜對(duì)照組相比明顯降低(P0.01),長時(shí)間歇組與持續(xù)訓(xùn)練組相比差異極顯著(P0.01);股四頭肌和腓腸肌MDA含量較安靜組相比,有下降的趨勢(shì),但差異性分析無統(tǒng)計(jì)學(xué)意義(P0.05)而比目魚肌MDA含量持續(xù)組與對(duì)照組相比顯著降低(P0.05),長時(shí)間歇組與對(duì)照組相比降低的趨勢(shì)極顯著(P0.01),且長時(shí)間歇組與持續(xù)組相比也呈現(xiàn)出顯著下降趨勢(shì)(P0.05)。2.SOD變化:運(yùn)動(dòng)組大鼠血清SOD的數(shù)值有上升的趨勢(shì),持續(xù)組與對(duì)照組相比差異性不顯著(P0.05)長時(shí)間歇組與對(duì)照組和持續(xù)組相比差異極顯著(P0.01)。心肌SOD值運(yùn)動(dòng)組與安靜組相比有升高的趨勢(shì),且長時(shí)間歇組與對(duì)照組相比有極顯著差異(P0.01),與持續(xù)組相比,也呈現(xiàn)出顯著差異(P0.05)。肝臟SOD值兩運(yùn)動(dòng)組與安靜對(duì)照組相比,有顯著性的差異(P0.01);長時(shí)間歇組與持續(xù)組相比差異極其顯著(P0.01)。股四頭肌和腓腸肌SOD含量兩運(yùn)動(dòng)組與對(duì)照組相比呈現(xiàn)上升趨勢(shì),但無統(tǒng)計(jì)學(xué)差異(P0.05),而比目魚肌SOD含量長時(shí)間歇組與對(duì)照組和持續(xù)組相比差異顯著(P0.05)。3.T-AOC變化:持續(xù)組和長時(shí)間歇組血清T-AOC活性與安靜組相比明顯升高,且差異性顯著(P0.05和P0.01);心肌T-AOC活性長時(shí)間歇組與安靜對(duì)照組和持續(xù)組相比有顯著差異(P0.01和P0.05),持續(xù)組與對(duì)照組相比無統(tǒng)計(jì)學(xué)差異(P0.05);肝臟T-AOC活性兩運(yùn)動(dòng)組與安靜組相比,活性明顯增強(qiáng),具有極顯著的差異(P0.01),且長時(shí)間歇組與持續(xù)組相比也具有顯著性差異(P0.05);股四頭肌和腓腸肌T-AOC活性與安靜組相比有所升高,但沒有顯著性差異(P0.05);比目魚肌T-AOC活性運(yùn)動(dòng)組與安靜組相比有明顯增強(qiáng)的趨勢(shì)(P0.05),持續(xù)組與長時(shí)間歇組相比無統(tǒng)計(jì)學(xué)意義(P0.05)。研究結(jié)論:(1)經(jīng)過6周持續(xù)與間歇跑臺(tái)訓(xùn)練后,兩種不同運(yùn)動(dòng)方式的大鼠體重增長幅度顯著低于安靜對(duì)照組,且長時(shí)間歇組顯著低于持續(xù)組,推測(cè)中等強(qiáng)度的長時(shí)間歇有氧運(yùn)動(dòng)有助于體重控制。(2)經(jīng)過6周持續(xù)與間歇跑臺(tái)運(yùn)動(dòng)訓(xùn)練后,持續(xù)組與長時(shí)間歇組血清,心肌,肝臟和骨骼肌自由基MDA的含量較對(duì)照組呈現(xiàn)下降趨勢(shì),兩運(yùn)動(dòng)組SOD和T-AOC的活性較安靜組均呈現(xiàn)增強(qiáng)趨勢(shì),提示適宜的運(yùn)動(dòng)有助于提高機(jī)體的抗氧化能力。(3)經(jīng)過6周持續(xù)與間歇跑臺(tái)訓(xùn)練后,長時(shí)間歇訓(xùn)練組比目魚肌SOD、T-AOC的活性顯著高于股四頭肌和腓腸肌,推測(cè)骨骼肌自由基的代謝可能與肌纖維類型有關(guān)。(4)在運(yùn)動(dòng)量相同的前提下,經(jīng)過6周持續(xù)與間歇跑臺(tái)訓(xùn)練后,長時(shí)間歇訓(xùn)練組血清和心肌自由基的代謝顯著優(yōu)于持續(xù)組和對(duì)照組,提示長時(shí)間歇訓(xùn)練組更利于自由基代謝。(5)在運(yùn)動(dòng)量相同的前提下,經(jīng)過6周持續(xù)與間歇跑臺(tái)訓(xùn)練后,長時(shí)間歇訓(xùn)練組肝臟自由基的代謝顯著優(yōu)于持續(xù)組和對(duì)照組,提示長時(shí)間歇訓(xùn)練有利于提高機(jī)體肝臟的解毒能力。
[Abstract]:Objective: To study the effects of 6-week continuous and intermittent treadmill exercise on the changes of MDA content and the activity of SOD, T-AOC in rat serum and different tissues (myocardium, liver, quadriceps femoris, gastrocnemius, and flounder). Methods:24 male SD rats were randomly divided into a quiet control group (S group), a continuous training group (C group) and a long-time rest training group (group I). The exercise group first carries out a 1-week adaptive run.
