8周間歇運(yùn)動(dòng)激活LIF-LIFR-STAT3信號(hào)和誘導(dǎo)骨骼肌細(xì)胞增殖
[Abstract]:Aim: to investigate the effects of intermittent exercise on the proliferation of skeletal muscle cells and the characterization of leukemia inhibitory factor (Leukemia inhibitory factor,LIF) and its downstream signal molecules in rats. Methods: three month old male SD rats were randomly divided into quiet control group (C), intermittent exercise group) and (CE), group (15 rats in each group). In the exercise group, 10 m/min 脳 10 min, followed by 25 m/min 脳 7 min and 15m/min 脳 3 min, 60 min/d,5 d / week 脳 8 weeks. The next day after exercise, the gastrocnemius muscle tissue was removed and weighed. The cross sectional area (cross-section area,CSA) of gastrocnemius muscle cells was measured by histological method. The expression of LIF and its receptor (Leukemia inhibitory factor receptor,LIFR, signal transduction and transcription activator (signal transducer and activator of transcription,STAT3 (signal transducer and activator of transcription,STAT3) and its phosphorylation level (p-STAT3) and proliferating cell nuclear antigen (proliferating cell nuclear antigen,PCNA) in gastrocnemius muscle were detected by Western blot. The expression of LIF,LIFR,PCNA and cell proliferation antigen marker (ki67) in gastrocnemius was detected by immunofluorescence, and the LIF/LIFR mRNA level in gastrocnemius was detected by RT-q PCR. Results: compared with group C, the weight of gastrocnemius muscle and the expression of LIF/LIFR gene and protein in gastrocnemius muscle increased significantly (P0.01), the expression of LIF/LIFR gene and protein increased significantly (P0.01), and the level of STAT3 phosphorylation increased significantly (P0.01) in CE group. The expression of PCNA and ki67 protein increased significantly (P0.01). Conclusion: 8 weeks intermittent exercise can promote the hypertrophy of skeletal muscle cells, activate the LIF-LIFR-STAT3 signal pathway and induce the proliferation of skeletal muscle cells.
