運(yùn)動(dòng)對(duì)大鼠骨骼肌細(xì)胞凋亡及鈣離子含量的影響

【摘要】:探討運(yùn)動(dòng)對(duì)大鼠骨骼肌細(xì)胞凋亡及鈣離子含量的影響。將大鼠分為對(duì)照組、一般運(yùn)動(dòng)組和強(qiáng)運(yùn)動(dòng)組,對(duì)照組大鼠正常活動(dòng),一般運(yùn)動(dòng)組大鼠跑臺(tái)速度為18m/min,強(qiáng)運(yùn)動(dòng)組大鼠跑臺(tái)速度為25m/min,一般運(yùn)動(dòng)組和強(qiáng)運(yùn)動(dòng)組大鼠均運(yùn)動(dòng)至力竭。分別檢測(cè)大鼠比目魚肌和脛骨前肌細(xì)胞凋亡情況,大鼠血清和大鼠后肢組織勻漿上清中超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量和鈣離子水平,大鼠肌肉組織中Bax、Bcl-2表達(dá)水平。結(jié)果發(fā)現(xiàn),一般運(yùn)動(dòng)組大鼠比目魚肌中細(xì)胞凋亡率和SOD活性均明顯高于對(duì)照組,差異極顯著(P0.01)。強(qiáng)運(yùn)動(dòng)組大鼠比目魚肌中的SOD活性明顯低于一般運(yùn)動(dòng)組,差異極顯著(P0.01)。試驗(yàn)組大鼠血清中MDA水平均明顯低于對(duì)照組,差異極顯著(P0.01)。一般運(yùn)動(dòng)組大鼠脛骨前肌和比目魚肌中MDA水平均明顯高于對(duì)照組和強(qiáng)運(yùn)動(dòng)組,差異極顯著(P0.01)。一般運(yùn)動(dòng)組大鼠比目魚肌中鈣離子水平和Bax表達(dá)水平明顯高于對(duì)照組,差異顯著(P0.05)。一般運(yùn)動(dòng)組大鼠比目魚肌中Bcl-2表達(dá)水平明顯低于對(duì)照組,差異極顯著(P0.01)。表明一般運(yùn)動(dòng)比高強(qiáng)度運(yùn)動(dòng)更易導(dǎo)致比目魚肌細(xì)胞凋亡,作用機(jī)制可能與鈣離子濃度、氧自由基水平、Bax、Bcl-2有關(guān)。
[Abstract]:To investigate the effect of exercise on apoptosis and calcium content of skeletal muscle cells in rats. The rats were divided into three groups: control group, general exercise group and strong exercise group. The normal activity of rats in the control group was 18 m / min in general exercise group, 25 m / min in strong exercise group, and exhaustion in general exercise group and strong exercise group. The apoptosis of rat soleus muscle and tibial anterior muscle, the activity of superoxide dismutase (SOD), the content of malondialdehyde (MDA) and MDAs and the level of calcium ion in the supernatant of rat serum and hind limb homogenate were measured respectively. The results showed that the apoptosis rate and SOD activity of soleus muscle in general exercise group were significantly higher than those in control group (P 0.01). The activity of SOD in the soleus muscle in the strong exercise group was significantly lower than that in the normal exercise group (P 0.01). The serum MDA levels in the experimental group were significantly lower than those in the control group (P 0.01). The levels of MDA in the anterior tibial muscle and soleus muscle in the general exercise group were significantly higher than those in the control group and the strong exercise group (P 0.01). The level of Ca ~ (2 +) and Bax expression in soleus muscle in general exercise group was significantly higher than that in control group (P 0.05). The expression of Bcl-2 in soleus muscle in general exercise group was significantly lower than that in control group (P 0.01). The results showed that general exercise was more likely to induce the apoptosis of soleus muscle cells than high-intensity exercise, and the mechanism might be related to the concentration of calcium ion and the level of oxygen free radicals (OFR) and BaxanBcl-2.
