噬菌體PF23生物學(xué)特性及其對(duì)小鼠肺炎克雷伯菌全身感染療效觀察

【摘要】:目的了解肺炎克雷伯菌裂解性噬菌體PF23的生物學(xué)特性,觀察其對(duì)小鼠全身感染的療效。方法電鏡觀察噬菌體PF23的形態(tài),一步生長(zhǎng)曲線以及對(duì)溫度和pH的敏感性;提取噬菌體基因組,酶切法鑒定其核酸類型,并初步估算其基因組大小;建立小鼠全身感染模型,觀察PF23的治療效果。結(jié)果 PF23的噬菌斑為有暈清晰透明圓斑,直徑約2mm;電鏡觀察PF23具有尾噬菌體目,長(zhǎng)尾病毒科病毒的形態(tài)特征;其在50℃的溫度下及pH 4~9范圍內(nèi)穩(wěn)定性好。一步生長(zhǎng)曲線顯示其潛伏期約為11min,裂解量約為41PFU/細(xì)胞。其基因組核酸不能被雙鏈DNA限制性內(nèi)切酶KpnI、EcoRI和HindIII切開,但能被BamHI切開,且酶片段大小合計(jì)27kb。用PF23治療由其宿主菌所致全身感染小鼠,小鼠的生存期均2d,7d存活率為37.5%;未治療對(duì)照組小鼠1d內(nèi)均死亡。結(jié)論肺炎克雷伯菌的噬菌體PF23潛伏期短,有較好的熱穩(wěn)定性和酸堿穩(wěn)定性,治療小鼠肺炎克雷伯菌全身感染效果顯著,值得進(jìn)一步研究。
[Abstract]:Objective to investigate the biological characteristics of lytic bacteriophage PF23 of Klebsiella pneumoniae and to observe its curative effect on systemic infection in mice. Methods the morphology, one-step growth curve and sensitivity to temperature and pH of bacteriophage PF23 were observed by electron microscope, the bacteriophage genome was extracted, the nucleic acid type was identified by enzyme digestion, and the genome size was estimated preliminarily. The model of systemic infection in mice was established and the therapeutic effect of PF23 was observed. Results the plaque of PF23 was halo clear and transparent round spot with a diameter of about 2 mm.
