CRISPR-Cas9技術(shù)在疾病治療中的應(yīng)用

【摘要】:CRISPR-Cas系統(tǒng)廣泛存在于細(xì)菌中,是一種高效、特異、操作簡(jiǎn)單易行、可對(duì)多種生物的基因組進(jìn)行遺傳改造的工具。與其他功能缺失的篩選技術(shù)(如RNAi、TALEN)相比,全基因組CRISPR-Cas9敲除技術(shù)顯示出更大的優(yōu)勢(shì)。這種技術(shù)被廣泛應(yīng)用于世界各地的實(shí)驗(yàn)室以及各個(gè)領(lǐng)域,尤其是在疾病研究中。利用CRISPR-Cas系統(tǒng),科學(xué)家在艾滋病、癌癥、亨廷頓氏病研究中取得了重大突破。本文將就CRISPR-Cas系統(tǒng)的最新進(jìn)展,尤其是在疾病研究中的最新應(yīng)用成果作一綜述,并對(duì)此技術(shù)在人類戰(zhàn)勝病魔的斗爭(zhēng)中將起到的關(guān)鍵作用進(jìn)行探討。
[Abstract]:CRISPR-Cas system, which exists widely in bacteria, is an efficient, specific, simple and easy-to-operate tool for genetic transformation of the genomes of a variety of organisms. Compared with other screening techniques (such as RNAi,TALEN), genome-wide CRISPR-Cas9 knockout shows more advantages. This technology has been widely used in laboratories all over the world as well as in various fields, especially in disease research. Using the CRISPR-Cas system, scientists have made major breakthroughs in AIDS, cancer, Huntington's disease. In this paper, the latest development of CRISPR-Cas system, especially the latest application results in disease research, are reviewed, and the key role of this technology in the fight against disease is discussed.
