【摘要】：目的:為評價康柏西普免疫原性,建立一種恒河猴血清中的抗康柏西普抗體檢測方法。方法:建立一種以康柏西普為包被蛋白、羊抗人Ig G Kappa light chain抗體為檢測抗體的ELISA檢測方法,并驗(yàn)證其精密度和專屬性。同時運(yùn)用細(xì)胞增殖實(shí)驗(yàn)確定陽性樣品抗康柏西普抗體的活性。結(jié)果:所建立的檢測方法最佳包被濃度為5μg·m L-1,檢測抗體的最佳稀釋倍數(shù)為2 500倍。陽性判斷閾(cut point)為0.162,批內(nèi)和批間變異系數(shù)均未超過15%�？蛋匚髌諠舛仍谛∮�10μg·m L-1時對檢測無干擾。24個血清樣品中,抗康柏西普抗體陽性率為62.5%,HUVEC細(xì)胞增殖實(shí)驗(yàn)顯示無陽性血清具有中和康柏西普生物學(xué)活性的能力。結(jié)論:經(jīng)驗(yàn)證,該方法可應(yīng)用于抗康柏西普抗體的檢測,為臨床前及臨床研究提供支持。
[Abstract]:Aim: to evaluate the immunogenicity of Compastep, a method for the detection of anti-Compastep antibody in rhesus monkey serum was established. Methods: to establish a ELISA method using Compastep as coated protein and goat anti-human Ig G Kappa light chain antibody as detection antibody, and to verify its precision and specificity. At the same time, cell proliferation test was used to determine the activity of anti-Compaxime antibody in positive samples. Results: the optimal concentration of coating was 5 渭 g 路mL ~ (- 1) and the dilution of antibody was 2 500 times. The positive judgment threshold (cut point) was 0.162, and the intra-and inter-assay coefficient of variation was not more than 15%. In 24 serum samples, the positive rate of anti-Compaxime antibody was 62.5%, while the concentration of Compastep was less than 10 渭 g 路mL-1. The positive rate of anti-Compastep antibody was 62.5% in 24 serum samples. HUVEC cell proliferation assay showed that the non-positive serum had the ability to neutralize the biological activity of Compastep. Conclusion: this method can be applied to the detection of anti-Compaxime antibody, which can provide support for pre-clinical and clinical research.
