TNF-α抑制劑對神經(jīng)病理性痛大鼠鎮(zhèn)痛作用及其機制探討

【摘要】:目的:探究腫瘤壞死因子α(tumor necrosis factorα,TNF-α)抑制劑XPro~汶1595對大鼠脊神經(jīng)結(jié)扎誘導的痛行為及脊髓背角白介素1β(IL-1β)、白介素6(IL-6)和TNF-α表達的影響。方法:運用L5脊神經(jīng)結(jié)扎的方法建立神經(jīng)病理性痛模型,通過鞘內(nèi)注射給予TNF-α抑制劑(XPro~汶1595)進行干預,采用Von Frey絲測定大鼠足底50%機械縮足反射閾值;運用Western Blot方法檢測大鼠脊髓背角IL-1β、IL-6和TNF-α的表達情況。結(jié)果:(1)行為學結(jié)果顯示:與對照組相比,脊神經(jīng)結(jié)扎后大鼠表現(xiàn)出明顯的機械性痛敏(P0.01),且可被鞘內(nèi)注射XPro~汶1595有效改善(P0.01);(2)Western Blot結(jié)果顯示:與對照組相比,脊神經(jīng)結(jié)扎組大鼠脊髓背角IL-1β、IL-6和TNF-α的表達顯著升高,且可被鞘內(nèi)注射XPro~汶1595明顯抑制(P0.01)。結(jié)論:鞘內(nèi)注射XPro~汶1595能夠顯著改善大鼠脊神經(jīng)結(jié)扎誘導的神經(jīng)病理性痛,其機制與抑制脊髓背角IL-1β、IL-6和TNF-α等炎性細胞因子的表達有關(guān)。
[Abstract]:Aim: to investigate the effects of tumor necrosis factor 偽 (TNF- 偽) inhibitor XPro~ Wen 1595 on the pain induced by spinal nerve ligation and the expression of interleukin 1 尾 (IL-1 尾), interleukin 6 (IL-6) and TNF- 偽 in the spinal dorsal horn of rats. Methods: the neuropathic pain model was established by ligation of L5 spinal nerve. TNF- 偽 inhibitor (XPro~ 1595) was injected intrathecally, and the reflex threshold of 50% mechanical foot contraction was measured by Von Frey wire. The expression of IL-1 尾, IL-6 and TNF- 偽 in spinal dorsal horn of rats was detected by Western Blot method. Results: (1) Behavioral results showed that compared with the control group, the spinal nerve ligation rats showed obvious mechanical pain sensitivity (P0.01), and the intrathecal injection of XPro~ Wen 1595 could be effectively improved (P0.01). (2) Western Blot results showed that the expression of IL-1 尾, IL-6 and TNF- 偽 in spinal dorsal horn of spinal cord ligation group was significantly higher than that in control group, and could be inhibited by intrathecal injection of XPro~ Wen 1595 (P0.01). Conclusion: intrathecal injection of XPro~ Wen 1595 can significantly improve the neuropathic pain induced by spinal nerve ligation in rats, and its mechanism is related to the inhibition of the expression of inflammatory cytokines such as IL-1 尾, IL-6 and TNF- 偽 in the dorsal horn of spinal cord.
