【摘要】：細(xì)胞培養(yǎng)作為定量檢測活病毒的金標(biāo)準(zhǔn),耗時(shí)長,通常需要數(shù)天到數(shù)周,光敏染料疊氮溴化丙錠聯(lián)合qPCR(PMA-qPCR)作為一種新型定量檢測病毒感染性的分析工具,具有快速便捷的優(yōu)點(diǎn)PMA預(yù)處理待測樣品,選擇性滲入滅活病毒衣殼,結(jié)合核酸后抑制其擴(kuò)增,使得僅活病毒核酸可以進(jìn)行qPCR本研究就PMA-qPCR檢測活病毒的機(jī)理、滅活病毒的方法進(jìn)行分析討論,并比較不同方案檢測病毒感染性的差異性。
[Abstract]:Cell culture, as a gold standard for quantitative detection of live viruses, takes a long time and usually takes several days to weeks. Guang Min dye azide and qPCR (PMA-qPCR) are used as a new analytical tool for quantitative detection of viral infection. PMA has the advantages of fast and convenient to pretreat the samples to be tested, selectively infiltrate the inactivated virus capsid, combine with nucleic acid to inhibit its amplification, so that only live virus nucleic acid can be carried out qPCR this study on the mechanism of PMA-qPCR detection of live virus. The methods of inactivation of virus were analyzed and discussed, and the differences of different methods for virus infection detection were compared.
