【摘要】：目的建立能夠同時(shí)檢測多重耐藥鮑曼不動桿菌(Multidrug-Resistant Acinetobacter Baumannii,MDRAB)多個耐藥基因的多重PCR反應(yīng)體系,了解MDRAB耐藥基因分布情況,為臨床治療及感染控制MDRAB提供分子生物學(xué)依據(jù)。方法針對MDRAB 4種耐藥基因blaoxa-23、bla Ade-B、blaint-1及bla ISCR-1分別設(shè)計(jì)4對引物,設(shè)計(jì)反應(yīng)條件在同一PCR反應(yīng)體系中同時(shí)檢測這4種耐藥基因,并采用建立的多重PCR反應(yīng)體系檢測臨床分離MDRAB耐藥基因攜帶情況。結(jié)果本研究成功建立了一個可以同時(shí)檢測4種MDRAB耐藥基因的多重PCR反應(yīng)體系,臨床分離68株MDRAB耐藥基因檢測顯示,1株同時(shí)攜帶有4種耐藥基因,5株攜帶有bla Ade-B、blaint-1及bla ISCR-1,62株攜帶有blaoxa-23和bla Ade-B。結(jié)論多重PCR可以快速、高效、低成本檢測MDRAB攜帶的耐藥基因,多重PCR檢測顯示blaoxa-23及bla Ade-B是MDRAB攜帶的主要耐藥基因。
[Abstract]:Objective to establish a multiplex PCR reaction system for simultaneous detection of multidrug resistant genes of Acinetobacter baumannii (Multidrug-Resistant Acinetobacter Baumannii,MDRAB) and to understand the distribution of MDRAB resistance genes in order to provide molecular biological basis for clinical treatment and infection control of MDRAB. Methods four pairs of primers were designed for blaoxa-23,bla Ade-B,blaint-1 and bla ISCR-1 of four resistant genes of MDRAB, and the reaction conditions were designed to detect the four resistant genes simultaneously in the same PCR reaction system. The multiplex PCR reaction system was used to detect the drug resistance genes carried by clinical isolates of MDRAB. Results in this study, a multiplex PCR reaction system was successfully established, which could simultaneously detect 4 MDRAB resistance genes. 68 clinical isolates of MDRAB resistance genes showed that 1 strain carried 4 drug resistance genes and 5 strains carried bla Ade-B,. Blaint-1 and bla ISCR-1,62 strains carry blaoxa-23 and bla Ade-B. Conclusion Multiplex PCR can be used to detect drug resistance genes carried by MDRAB quickly, efficiently and at low cost. Multiplex PCR analysis shows that blaoxa-23 and bla Ade-B are the major drug resistant genes carried by MDRAB.
